In addition, the preventive effects of inosine and Rhizoma Ligustici Chuanxiong on ischemia and reperfusion cell injury were studied. The results showed that the preventive effects of inosine and Rhizoma Ligustici Chuanxiong were obvious. Redistribution and slight increase of calcium occurred during ischemic period in the proximal tubule cells. In this period the changes of plasma membrane permeability were gentle. Following the extention of reflow time, a large amount of calcium deposited in the cells and mitochondria. The plasma membrane became broken.

本实验采用钙离子细胞化学定位法、膜通透性细胞化学示踪法结合普通超微结构及EDX电镜分析,从细胞水平观察了肾近端小管上皮细胞缺血损伤情况以及药物对它的影响。实验结果表明:肌苷、川芎对细胞缺血损伤有保护作用。缺血期,细胞中及线粒体内钙离子增多。此时,质膜的损伤并不严重。缺血后复流,随着复流时间的延长,细胞中及线粒体内钙离子进行性增多,质膜的损伤也越来越重。线粒体内钙离子含量与胞质中钙离子含量有密切关系。

采用庆大霉素(GM)肾毒性大鼠模型,探讨肾皮质线粒体、溶酶体膜流动性与其膜脂质过氧化(SOD)的关系。结果表明GM组0.5h,线粒体、溶酶体膜荧光偏振度、微粘度和各向异性值非常显著地升高(P<0.01),亚细胞器悬液丙二醛(MDA)含量亦显著升高(P<0.05),并与膜微粘度升高呈非常显著正相关(P<0.001)。GM组1h,肾近端小管上皮细胞线粒体肿胀、嵴断裂、消失,溶酶体数量增多和体积增大,并有髓样小体形成。提示GM肾中毒时肾皮质细胞器膜流动性下降,可能与膜脂质过氧化反应增强有关;并在线粒体及溶酶体损伤中可能具有重要意义。
