This paper is the first report of the expression of Antheraea yamamai fibroin gene in Bombyx mori. We have constructed an yeast artificial chromosome (YAC) clone containing the fibroin gene of A. yamamai, and transferred this DNA clone into the fertil- ized eggs of B. mori. Dot blot indicates that the fibroin gene has integrated into the genome of B. mori. By analyzing the fibroin amino acid composition and the silk solubility, we found that A. yamamai fibroin gene has expressed in some of the transgenic B....

This paper is the first report of the expression of Antheraea yamamai fibroin gene in Bombyx mori. We have constructed an yeast artificial chromosome (YAC) clone containing the fibroin gene of A. yamamai, and transferred this DNA clone into the fertil- ized eggs of B. mori. Dot blot indicates that the fibroin gene has integrated into the genome of B. mori. By analyzing the fibroin amino acid composition and the silk solubility, we found that A. yamamai fibroin gene has expressed in some of the transgenic B. mori. The existence of YAC sequence in the F, progeny of transgenic B. mori suggested that YAC is a medium for the transfer of fibroin gene. There is only one copy of A. yamamai firoin gene in the haploid of F, progeny.

本文为天蚕Antheraeyamamai丝素基因在家蚕Bombyxmori成功表达的首次报道。我们构建了天蚕丝素基因的YAC克隆,然后把含有该克隆的DNA溶液导入家蚕受精卵。分子杂交实验证明天蚕丝素基因已整合到家蚕基因组中。通过丝心蛋白氨基酸组分分析以及茧丝的溶解性比较,发现有部分转基因家委表达了天蚕丝素基因。F2代的转基因家蚕蛾的染色体DNA中同时还存在YAC序列,说明YAC对丝素基因具有介导作用。天蚕丝素基因以单拷贝形式存在于转基因家蚕中。

In order to study the effect of transposon K1.4 on foreign genes' integration frequency, a recombinant plasmid SKFL was constructed which contained Fb(the 5'-flanking region from -850bp to +75 of the fibroin heavy chain of Bombyx mori), transposon K1.4 and luciferase gene (Luc), used as a report gene. The other recombinant plasmid pFL containing Fb and Luc was constructed as control. The fresh Bombyx mori eggs laid in 3-8 hous were divided into two groups: one was transferred with SKFL as an anexperimental...

In order to study the effect of transposon K1.4 on foreign genes' integration frequency, a recombinant plasmid SKFL was constructed which contained Fb(the 5'-flanking region from -850bp to +75 of the fibroin heavy chain of Bombyx mori), transposon K1.4 and luciferase gene (Luc), used as a report gene. The other recombinant plasmid pFL containing Fb and Luc was constructed as control. The fresh Bombyx mori eggs laid in 3-8 hous were divided into two groups: one was transferred with SKFL as an anexperimental group and the other was transferred with pFL as control group. After the surviving eggs were raised to the fifth instar larvae, DNAs, RNAs from the silkglands and the bodies of silkworms were extracted respectively. DNA Dot Blot and Southern Blot indicated that luciferase gene had been integrated into the genome of Bombyx mori in the experimental group, but the integration sites of luciferase gene distributed randomly, which accorded with the mosaicism of the foreign genes* integration in host cells. However, luciferase gene did not integrate in the genome of Bombyx mori in the control group. These results provided us with the evidence that K1.4 played an important role in increasing the integration frequency of the foreign gene. Moreover, the negative result of RNA Dot Blot indiated that luciferase gene did not express in the fifth instar larvae of Bombyx mori. Nevertheless, the transposon K1.4 could be a very useful integration vector in constructing transgenic silkworms.

合适的整合表达载体对于获得转基因家蚕具有重要意义,为此本文构建了包含家蚕转座子K1.4、丝素基因上游启动子序列和荧光素酶基因cDNA的质粒SKFL以及不含K1.4的质粒pFL,并将其通过扎卵法转移到家蚕受精卵中.通过对当代转基因家蚕5龄幼虫丝腺和蚕体DNA进行斑点杂交和Southern杂交发现,荧光素酶基因在家蚕不同组织中的整合是随机的,但携带K1.4的质粒SKFL的整合频率明显高于质粒pFL.这说明转座子K1.4可以有效提高外源基因在转基因家蚕中的整合频率,从而有可能成为构建转基因家蚕的一种有用的整合载体.

将线性化的含核酶基因的重组质粒pGL2Rz用基因枪导入家蚕早期受精卵中(G0代)。检测了G1代五龄蚕血淋巴的荧光素酶活性,并且从G2代开始通过家蚕核多角体病毒(NPV)攻击来筛选抗性较高的家蚕进行传代。在G4代获得一区NPV抗性强度比对照组高10倍的转基因家蚕。对该区五龄蚕基因组DNA的PCR检测和Southern杂交证明,核酶基因多拷贝地整合在家蚕染色体上。利用RTPCR能检测到蛹期核酶的表达。研究结果表明已经获得了抗NPV的核酶转基因蚕。
