Four hundred and twenty five fertilized eggs, which were injected with pure 24 kb Ren-2 gene, were implanted into the oviducts of 22 pseudopregnant recipients in a dose about 20 eggs per female. Transgenic animal technology is widely used in pharmacology and toxicology research. Up to now, many transgenic animal models for complex diseases such as Alzheimers disease, insulin dependent diabetes mellitus, hepatitis B virus infection and human immunodeficiency virus infection have been established for drug screening and evaluation. In addition, mice lacking 5 HT receptor, hypertension rats, interferon γ gene disrupted mice and P glycoprotein knockout mice have been developed for respectively studying the pharmacological profiles of 5 HT, renin, IL 12 and P glycoprotein. The applications of transgenic animals in toxicology include experiments on mutagenesis, carcinogenesis, DNA damage and repair, risk assessment and illumination of toxicity mechanism of chemical carcinogenesis and so on.

转基因动物技术在药理学和毒理学研究中的应用有十分广泛的前景,目前已建立了早老性痴呆、胰岛素依赖性糖尿病、乙型肝炎病毒感染和人类免疫缺陷病毒感染等多种复杂疾病的转基因动物模型,用于新药的筛选和评价。此外,还制备了5-羟色胺受体缺陷小鼠、高血压转基因大鼠、γ干扰素基因和多药耐药基因缺陷转基因小鼠,分别用于5-羟色胺受体、肾素、白介素12和多药耐药蛋白的药理特性研究。在毒理学中的应用包括致突变试验、致癌试验、DNA损伤与修复、化学致癌危险度评定和毒理学机制的研究等。

Conclusion This paper suggested that HLA-B_27 gene has evident correlationwith AS and this study has laid a foundation for the further study of the etiology, pathology and therapy of AS.

目的构建带HLA-B_27基因的转基因大鼠,进一步研究HLA-B_27与强直性脊柱炎(AS)的发病原因及其机理。方法应用转基因技术,把已克隆的人类AS基因HLA-B_27基因组DNA导人Wistar大鼠受精卵中,并移植到假孕母鼠输卵管内,21d后,出生27只原代大鼠。结果经过PCR检测阳性后,再继续进行Southern印迹分析,确认3只大鼠为带HLA-B_27基因转基因大鼠,整合率为11.11%,其中1只转基因大鼠出现外周关节炎及脊柱强直表现。结论本研究进一步表明HLA-B_27与AS的强相关性,为研究AS病因病理及治疗打下了基础。
