Fatty acid and steroid nitroxide free radicals have become valuable tools for investigating membrane structure and function. In this paper we describe an improved procedure for the synthesis of the 5'-nitroxide methyl palmitare (Ⅰ) and a new compound, nitroxide progesterone (Ⅱ). The improved methods gave high yields of the free radical products and reduced reaction times. These products have been shown to be good in quality by means of elementary analysis, thin-layer chromatography, infrared and mass spectra.The orientation of the nitroxide-fatty acid and nitroxide-steroid spin labels on membranes are different from each other. The former nitroxide radical axis (2Pπ orbital) is perpendicalar to the membrane surface while the latter is parallel to it. When the nitroxide compounds Ⅰ and Ⅱ were used to spin label a liposome membrane and the lung cell (normal and cancerous) membrane of the Chinese hamster, the electron spin resonance (ESR) spectra could afford information regarding the mobility of the probe, the polarity of the enviroment and the microscopical viscosity. It was found that in the low magnetic field region the ratios of anisettopic h_((a)) and h_((1)) spectra showed a notable variation. There was a linear correlation between the anisotropio motion M and the rotational correlation time τ_v. Thus, we proposed the formula:

In this paper the effect has been studied of some antitumour drugs(thioproline,cytosine,actinomycin and 5-fluoro-2′-deoxyuridine) acting at different levels onthe membrane fluidity of the normal Chinese hamster lung cell V_(79) and cancer cellV_(79)-B_1.From the spectra obtained the order parameter,mean fluctuation angle,rotational correlation time and microviscosity of cell membrane fatty acid chainbefore and after treatment with antitumour drugs were calculated.It was found thatafter treatment with these drugs the order parameters were raised,the mean fluctua-tion angles were lessened and the rotational correlation times and micro-scopicviscosities were raised,thus lowering cell membrane fluidity.The effects of theseantitumour drugs on tho hy drophobic parts of the fatty acid chain are similar,butthe effect of thioprolino on the surface of the membrane is the largest,while thoseof cytosine,actinomycin and 5-fluoro-2′-deoxyuridine are smaller.The kineticcurves of the order parameters corresponded to the times of the antitumour drugtreatment,were madeFrom these curves it was found that the response of V_(79)-B_1 tothe drugs was faster than V_(79) except for actinomycin.

本文用脂肪酸自旋标记Ⅲ(10,3)在ESR波谱仪上研究了作用于不同环节的抗癌药物硫杂脯氨酸、阿糖胞苷、放线菌素D和5-氟-2′脱氧尿嘧啶对中国地鼠肺正常细胞V_(79)和癌变细胞V_(79)-B_1膜流动性的影响。从所得的ESR波谱计算了抗癌药物处理前后细胞膜脂肪酸链的序参数、平均涨落角度、旋转相关时间和微观粘度的变化。发现用抗癌药物处理以后,V_(79)和V_(79)-B_1细胞膜脂肪酸链的序参数增加,平均涨落角度变小,旋转相关时间和微观粘度增大,细胞膜流动性变小。这四种药物对细胞膜的疏水脂肪酸链作用类似,而硫杂脯氨酸对细胞膜表面作用最大,阿糖胞苷,放线菌素D和5-氟-2′脱氧尿嘧啶依次减小。做了序参数随药物处理时间的动力学曲线。发现除放线菌素D外,V_(79)-B_1细胞膜序参数对其他三种药物的反映比V_(79)细胞膜快。
