Simultaneously some ultrastructural changes including mitochondrial swelling, atrophy of Golgi complex, decreasing in pinocytotic vesicles and stereocilia were found in principal cells of epididymis within 3 days after medication.In this experiment the animals received 3, 6 and 9 courses of treatment respectively, each course included two days of administration with a daily dose mentioned above and eight days of cessation of medication.The specimens of initial segment of epididymis, liver and kidney were taken at the end of 3 rd, 6 th and 9th courses of treatment for electron microscopy.The principal cells of initial segment of epididymis revealed a normal feature at the end of 3 rd and 9 th course of treatment. But at the end of 6 th course of treatment, the atrophy of Golgi complex and the decrease in stereocilia and pinocytotic vesicles were found in some principal cells.Any ultrastructural changs of sperms in the initial segment were not found by electron microscope.There was also mitochondrial changes with striking increase in the density of the matrix and reduction of size in some liver cells at the end of 6 th course of treatment, but not at the end of 3 rd or 9 th course of treatment. The epithelial cells of nephron in all the specimens revealed a normal ultrastructural feature.The results suggest that the ultrastructural changes of the principal cells and the liver cells induced by the mixture of the drugs could be recovered at the end of treatments.

以每日5mg/kg的3-氯丙二醇及75mg/kg的2,3-氧丙醇同时给大鼠灌胃,连续给药2天后停药8天,共10天为一疗程。观察了3,6,9疗程结束时,附睾起始部主细胞的超微结构改变,及附睾管起始部的精子和肝脏、肾脏的超微结构。发现在3、6疗程末,附睾主细胞的高尔基复合体扁平囊变窄,大泡皱缩。在6疗程末,细胞表面的吞饮小泡和静纤毛减少。仅在3疗程见到粗面内质网扩张。9疗程的超微结构与对照比较无明显区别。结果提示,每一疗程药物引起的附睾主细胞超微结构的改变,在多数动物是可恢复的。但由于个体差异,部分动物于疗程之末,尚未完全恢复。在第6疗程末的肝脏,有部分肝细胞滑面内质网扩张,线粒体体积缩小,基质电子密度增高。这些变化可能是一种轻度的细胞损伤,这种损伤在3,9疗程均未发现,说明它与疗程长短不呈平行关系。肾尿细管上皮细胞及附睾起始部精子的超微结构,无可见改变。这一结果为阐明3-氯丙二醇及2,3-氧丙醇合并用药的抗生育作用,及对有关内脏的毒性作用,提供了形态学的依据。

There was highly Positive correlation between the values of pH, BB and HCO_3 in urine and the different average accumulative intake of overdue tetracycline. All the epithelial cells of the renal Proximal tubules became cloudy swelling and the cavity narrow in experimental group 4 and 5.Electron microscopy disclosed that the mitoehondria were swelling or its cristae disappearance in group 1 and 2. We also found that the effective Phase of overdue tetracycline to dameage mitochonrial ultrastructure was different from mercury bichloride. On the basis of the results,the authors believe that the effect of overdue tetracycline on the mitochondiral damage may be as starting factor causing injury to the kidney.

本文用毒理学研究手段,探讨用过期四环素致肾脏损害的动物模型的制作方法及观察手段,为进一步研究肾脏损害,特别是肾小管酸中毒的发病机理,病理改变、临床表现、诊断及治疗打下基础。本项研究用过期四环素针(粉)剂,以每日0.5mg/10g、每日1mg/10g、每日2mg/10g的剂量给小鼠灌胃,按不同累积摄入量分5个组。观察指标有:血气分析,尿气分析,病理及超微结构的变化等。通过形态学观察证实,过期四环素能制出肾脏近曲小管损害的动物模型,初步观察到过期四环素对肾脏超微结构效应时相,并提出小鼠的血气分析、尿气分析资料。
