The toxic effect of cyclophosphamide on the proliferative cell population of hair follicles plucked from the human scalp was examined by the in vivo nuclear aberration assay.
      
The observed increase in nuclear aberrations following low dose as well as high dose cyclophosphamide suggests that it is feasible to use the nuclear aberration assay for in vivo human genotoxicity testing, using proliferating hair follicle cells.
      


Wistar rats (170~215g), male 60 and female 60, supplied by the Lab Center of Chinese Academy of Medicine, were given orally 2,4, 6-trinitrotoluene(TNT) suspension in 2.5% starch(1ml/100g wt) and observed for 10 days; placebo given to control groups. Liver samp les were taken for HE and ultrastructure exams. Results suggest that TNT LD_(60) in rats was 1663.8 mg/kg(95% CI 1990.9~1328. 6mg/kg) for female and 1837.8mg/kg(2163.5~1561.3mg/kg) for male. Besides common liver damages, cholangiolar cholestasis and nuclear...

Besides common liver damages, cholangiolar cholestasis and nuclear deformity of hepatic cells were found in 50% of the lower-TNT-dosepoisoned rats. These findings are in conformity to the theory that TNT may be an allergenic and carcinogenic substance to some people.

本实验取Wistar大鼠雄、雌各60只,用悬浮于2.5%淀粉液内的TNT研碎,以1ml/100g灌胃,一次染毒后观察10天,死亡鼠随即取肝。至第11天中毒未死及对照组鼠全部处理并取肝作光镜和超微结构观察。结果表明TNT对雌鼠LD_(50)为1663.8mg/kg发现,雄鼠为1837.8mg/kg。肝脏除发现变性、坏死及线粒体破坏外,低剂量中毒鼠约50%出现胆小管淤胆和肝细胞核畸变。从而提示TNT可能与变态反应以至致癌作用有关。

应用Ag-NoRs染色技术及电镜观察,对48例骨巨细胞瘤各个级别的基质细胞,多核巨细胞进行了分析,以揭示其细胞增生活跃,结果表明Ag-NoRs计数除Ⅱ─Ⅲ级之间基质细胞无显著性外(P>0.05)其它各级之间均有高度显著性,电镜下分化差的骨巨细胞核畸变显著、内质网极度不规则并呈迷宫样,多核巨细胞随着级别增大,其数量减少。Ag-NoRs在Ⅰ─Ⅱ级GCT之间无明显差异(P>0.05)而面积有高度显著性。其他各级之间Ag-NoRs和面积均有高度显著性。通过Ag-NoRs和电镜观察可反应出各级之间细胞增生活跃的关系,可作为判断良、恶性的指标,指导临床手术治疗,预测予后复发机会
