Based on the practices of leather manufacturing,it was suggested that the activity ofcollagenolytic enzymes should be its capability of hydrolyzing and solubilizating hidecollagen into soluble fiber.A novel and rapid assaying method for the collagenfiberhydrolyzing proteases was then developed against insoluble acid-relaxed collagenfiber la-belled with reactive dye.Its activity was calculated by the color of soluble hydrolysate byspectrophotometry.This assaying method meight been used for the bating enzymes.

Zn~(2+) was the necessary metal ion. It could be completely inhibited by EDTA. Its hydrolysis against collagen was mainly characteristic of histolysis of collagen fibre. The hydrolyzing rate of the collagenolylic protease against denatured collagen was ten times more than against natural collagen.

枯草杆菌AS1.398蛋白酶制剂广泛用于制革工业中的脱毛和软化。本研究采用铬皮粉亲和层析等方法分离纯化了其中的胶原水解酶,比活力达到380U/mg。初步确定其分子量<10000,等电点<3.5,最佳反应条件为pH10,T=50℃,Zn~(2+)为必需离子,可被EDTA完全抑制,对胶原的水解作用显著地表现为纤维组织的溶解,对变性胶原的水解速度为天然胶原的10倍以上。
