The study on the technical conditions of beer fermentation with high temperature and press-are were carried out.The optimal technology was presented.The analytical results of the qualities indicated that there was not a very significant difference between the beer thus obtained and that obtained by the conventional fermentation method.The use of the technology of the high temperature and pressure fermentation can reduce fermentationtime by about 65% and multiply the equipment capacity.

Pullulan, a kind of exopolysaccharides produced by Aureobasidium pullulans, has a number of favorable performances such as film-forming, high adhesion, low digestibility, nontoxicity etc., and is widely used in food, pharmaceutical, chemical industry, petrochemistry, light industry and other manufacture industries. Therefore, it has been paid great attention by scientists and enterprisers. In this paper, two Pullulan mutant strains, ZY047 and ZY073, with low pigment content and high pullulan of 1515 mg/mL and 14.22 mg/mL respectively, were obtained by UV mutagesis. The fermentation status was greatly improved and a lot of swollen cells and chlamysdospores were observed. Through adjusting the C/N ratio of culture medium and fermentation conditions,an optimum culture medium has been obtained with 5% sucrose , 0.5% KH2PO4, 0.1% NaCl, 0.02% MgSO4·7H2O , 0.03% (NH4)2 SO4 and 0.09% of yeast extract. The optimum fermentation conditions were as follows: initial pH value-5.5, liquid culture age-48 h of the second-generation, inoculated volume-5% and culture flask volume-25%. The pullulan yield of mutant strain ZY047 was 21.11 mg/mL and its conversion rate was 54.10% under the optimized conditions, which was 51% and 32% higher than that under the original conditions.

本研究应用紫外(UV)随机诱变获得了两株多糖产量高、色素低的出芽短梗霉突变株ZY047和ZY073,其产量分别为15.15和14.22mg/mL。发酵状况大大改善,产生大量的膨大细胞和厚垣孢子。通过短梗霉的培养基适宜碳氮比和发酵条件优化,获得优化的培养基方案为:蔗糖5%,KH2PO40.5%,NaCl0.1%,MgSO4·7H2O0.02%,(NH4)2SO40.03%,酵母浸出粉0.09%。优化的发酵条件为:初始pH65,接种量5%,菌龄48h的二级种子,装瓶量75mL,占总容量的25%。采用上述优化的发酵培养基及发酵条件,应用ZY047突变株验证获得短梗霉多糖产量21.11mg/mL,转化率达54.1%。与原发酵条件作对照相比,多糖产量和转化率分别提高51%和32%。
