采用RFLP技术,对四川白鹅和朗德鹅及其杂交后代进行了线粒体DNA(mtDNA)多态分析。在所使用的19种限制性内切酶中,有4个酶(EcoRV、HaeⅡ、HincⅡ和KpnⅠ)检测出多态,共获得两种mtDNA单倍型,四川白鹅和以四川白鹅为母本杂交后代的mtDNA为Ⅰ型,朗德鹅的mtDNA为Ⅱ型。以上结果为中国鹅和欧洲鹅存在两种不同起源学说提供了分子遗传学的证据。EcoRV、HaeⅡ、HincⅡ和KpnⅠ四种限制性内切酶的两种限制性态型可以作为鉴别两种不同起源家鹅的母系分子遗传标记。线粒体DNA在家鹅中也遵循严格的母系遗传。

Mitochondrial cytochrome b(Cyt-b) sequence were used to study the molecular evolution of Anser cygnoides (15 breed of Chinese goose),Anser anser(2 breed of domestic European goose) and Anser albifrons(the lesser white-fronted goose, Genbank accession number AF363031), insertion/deletion or frameshift mutation were not found in Cyt -b gene sequences (1143bp),the average rates of synonymous (dS) and nonsynonymous substitution (dN) were 0.0787 and 0.0011 respectively .there was a significant difference between dS and dN (Z=4.713,P<0.01).Because dS<0.5,it was used to constructe the phylogenetic tree.The phylogenetic trees supported the view of the double origin of Chinese goose, which meaned that domestic Chinese goose (excluding Yili) was derived from Anser cygnoides and domestic European goose(Landoise,Rhin), and Yili was derived from Anser anser.

采用PCR和DNA测序技术测定了6个中国家鹅品种和2个欧洲鹅品种25个个体线粒体DNA(mito-chondrial DNA,mtDNA)的部分序列,经测序分析表明该序列(1 401 bp)包括了细胞色素b(cytochrome b,Cytb)基因的完整序列(1 143 bp)。通过对家鹅线粒体基因组的研究,首次报道了家鹅线粒体Cytb的跨膜螺旋结构,并对家鹅Cytb跨膜螺旋结构及各区域序列的变异特征进行了分析。结果表明:鸿雁、灰雁和白额雁的Cytb具有完整而且完全相同的跨膜螺旋结构;家鹅Cytb基因的核苷酸序列在膜内、膜间和膜外的变异位点数分别为12、9和10,膜内、膜间的变异均属同义替换,而膜外有2个位点发生了非同义替换,表明跨膜螺旋的膜外区域变异程度高于膜间和膜内。研究的结果将为进一步探讨家鹅细胞色素b结构与功能的关系奠定基础。所测定的基因序列已登录国际GenBank数据库,序列号为AY427800~AY427805和AY427812~AY427814。
