Peroxidase-antiperoxidase (PAP) technique was used to demonstrate the distribution of leuenkephalin-like immunoreactivity in the cells of the adrenal medulla and celiac ganglia of guinea pigs. Almost all of the chromaffin cells in the adrenal medulla exhibited the immunoreactivity. The immunoreactivity was also found to be present in a few of small intensely fluorescent cells and some nerve fibers while the principal neurons in the celiac ganglia did not show the immunoactivity.After the histofluorescence reaction induced by the trihydroxyl iodine most of chromaffin cells, if not all, displayed the fluorescence characteristic of adrenaline fluorophore.The results from both the immunoreaction and the fluorescence histochemistry have been discussed in the light of their interrelationship. It is suggested that the leu-enkephalin-like immunoactivity is associated only with the adrenalin-containing cells of sympatho-adrenal system.
