The two-way data is usually necessary for the quantitative analysis of the gray system. A new way to the two-way kinetic-spectral data as the data source for the quantitative analysis of the gray system has been proposed by monitoring the chemical reaction multiwavelength. According to the character of the two-way kinetic-spectral data, the constrained background bilinerization algorithm was used to resolve the ana-lyte in the gray system. Simulated data has validated the liability of this method. A kinetic-spectral matrix was collected during the process of the alkaline hydrolysis of the diethyl phthalate (DEP), where ethyl benzoate was added deliberately as the unknown background. By the method in this paper, the quantitative analysis for the DEP was well done though the kinetic spectra and the pure spectra of the background and the analyte overlapped seriously.

Determination of pigments from tobacco leaf was studied by microcolumn high performance liquid chromatography. The tobacco leaf pigments was extracted from tobacco leaf with 90% acetone and then purified and enriched by solid phase extraction with Sep-Pak-C18 cartridge. Chlorophylls-a and b, (-carotene, lutin, neoxanthin, violaxanthin, pheophorbide and other tobacco leaf pigments could be separated and determinated on a Waters Xterra TM RP18 (1.0×50 mm, 2.5 μm) microcolumn, eluting with methanol-isopropyl alcohol-water...

Chlorophylls-a and b, (-carotene, lutin, neoxanthin, violaxanthin, pheophorbide and other tobacco leaf pigments could be separated and determinated on a Waters Xterra TM RP18 (1.0×50 mm, 2.5 μm) microcolumn, eluting with methanol-isopropyl alcohol-water gradient (1+1). Recovery of tobacco leaf pigments were 95%~105%, with relative standard deviation of 1.6~2.5%. The method could be applied to determine pigments from tobacco leaf with satisfactory result.

通过微柱高效液相色谱法测定烟草样品中的植物色素;用 90%丙酮震荡浸提烟草样品中的色素,提取液经WatersSep Park-C18固相萃取小柱预分离和富集,以WatersXterraTMRP18 (1. 0×50mm, 2. 5μm)色谱柱为固定相, (1+1)甲醇异丙醇溶液-水梯度为流动相,烟叶中的叶绿素 a、叶绿素 b、β 胡萝卜素、叶黄素、新叶黄素、紫黄质和脱镁叶绿素等色素分别在其最大吸收波长监测下分离提取,色谱峰面积定量。该方法标准回收率95% ~105%, RSD为 1. 6% ~2. 5%。将其用于烟草中天然色素含量的测定,结果令人满意。
