In this study,the karyotype and Giemsa banding pattern of the chromosomes of cultured peripheral blood lymphocytes in R.r.roxellanae were investigated.The chromosome number of this species is 44 in both sexes.In R.r.raxellanae,as in other monkeys,sex is determined by specific sex chromosomes,i.e.the male is XY and the female is XX.The 21 pairs of autosomes consist of 7 pairs of metacentric chromosomes,13 pairs of submetacentric ones,1 pair of acrocentric ones.Chromosome measurements were made from highly enlarged photographic prints.Table II lists the results obtained including the relative length,arm ratio and centromere index of each chromosome.Both chromosomal and the chromatid aberrations were observed.They were 0.67% and 2%,respectively.Finally,Giemsa banding pattern analysis of chromosomes of R.r.roxellanae were carried out.The results show that each homologous pair has its own special banding pattern,so that each of them is easily recognizable.Idiograms of chromosome complements with the Giemsa banding pattern were constructed.

The chromosomes of amphibia, because of their large size and a small number are very well suited for cytogenetic studies. Amphibian embryos are favorite material in the study of differentiation and the differentiation phenomenon will eventually be analyzed at the chromosomal level. In this experiment, seven females and ten males of Cynops orientalis and four females and three males of Pachytriton brevipes were used. These animals were collected from Zhejiang, Jiangsu, and Anhui provinces of our country.

1.东方蝾螈(Cynops orientalis)、肥螈(Pachytriton brevipes)染色体数均为2n=24。按染色体相对长度、臂比、着丝点指数可将12对染色体分成三组。染色体1—4为第一组,均为中部着丝点染色体。染色体5—8为第二组,除染色体7为亚中部着丝点外,其余为中部着丝点染色体。染色体9—12为第三组,除染色体9为中部着丝点外,其余都为亚中部着丝点。东方蝾螈和肥螈染色体核型相似,但却各具独特的C-带带谱图式。 2.经间接免疫过氧化物酶染色后,在东方蝾螈和肥螈肠上皮细胞染色体的着丝点两侧和长、短臂的端粒部位都显示棕褐色的5-甲基胞嘧啶特异位置的深着色,染色体的长臂、短臂上则为浅棕色。 3.东方蝾螈和肥螈雌雄个体肠上皮细胞的染色体核型中均未见染色体的异型性配对。

Chromosomal preparations were made from cultured embryonic cells and peripheral blood leucocytes. Some interesting facts have been found in our experiment :1. The diploid chromosome numbers of two species and their intergeneric hybrid are the same (2n=78). But their total numbers of chromosome arms are different [N. P.: Muscovy duck-82; Ldancheng duck-84 (cf) or 83 (?); F1-83 (?) or 82 (?)].2. Both the centromeric index and arm ratio of the 1st and 2nd pair of chromosomes in hybrid show that they are the intermediate type of their parents.3. One of the Z-chromosome of the male hybrid is a subtelocentric chromosome with an obviously short arm, while the other is an actocentric one without a short arm. It is evidently shown that they are descendants from different parents. This contributes much to confirm the Mendel's law of segregation by means of the characteristic variance of centromeric position of sexual chromosome and rectify the authenticity of hybrid of descendant from different species.

采用胚胎组织细胞和外周血白细胞短期培养法,分析了番鸭、连城鸭及其属间杂种F_1的体细胞染色体核型。番鸭的染色体2n=78,连城鸭2n=78±。它们的第1、2对常染色体分别为亚中或中间着丝点,其余均为近端着丝点;性染色体ZZ—ZW按大小顺序排列,Z为4,W为7—8。番鸭的Z和W染色体均属近端着丝点;连城鸭的Z染色体具明显短臂,属亚端着丝点,W染色体不具短臂,属近端着丝点,杂种F_1与亲本比较,染色体数目相同,但总臂数不同;第1、2对常染色体着丝点指数和臂比介于两亲本之间,其两个同源姐妹染色体的臂比分别偏向于父本和母本;性染色体着丝点位置具备两亲本的特点,可作为鉴定属间杂种真实性的依据。
