In this study, the karyotype of Fasciolopsis buski has been investigated by the air drying method. The results show that the diploid number of chromosomes is 2n =14. The means and standard deviations of the relative lengths, arm indices and centromeric indices of the chromosomes have been calculated. In this study, the karyotype of Fasciolopsis buski has been investigated by the air drying method. The results show that the diploid number of chromosomes is 2n =14. The means and standard deviations of the relative lengths, arm indices and centromeric indices of the chromosomes have been calculated. According to the nomenclature recommended by Levan et al (1964), the karyotype of F. buski obtained consists of one pair of large-sized metacentrics (m), 2 pairs of medium-sized metacentrics (m), one pair of medium-sized submetacentrics (sm), one pair of small-sized metacentrics (m), one pair of small-sized submetacentrics (sm) and one pair of small-sized telocentrics (t).Idiogram of chromosomes has been constructed from measurements of 19 good metaphase spreads in mitoses.The karyotype of F. buski of this paper is compared with that reported by Gao et al (1952). Statistically, there are very distinct differences (p<0.01) of the means of the arm indices and centromeric indices of the 3rd, 4th and 6th pairs of chromosomes between the two results. Basing on the arm indices and centromeric indices of these chromosomes calculated by us, the 4th and 6th pairs of chromosomes are submetacentrics while Gao et al (1952) considered that these 2 pairs of chromosomes are all metacentrics

Objective To compare the differences on morphology and chromosome mitotic karyotypes between An. minimus A and An. minimus C to find the differential characters between two groups. Methods The Anopheles samples collected in the field were examined by morphological method, and those classified as An. minimus were brought back to the lab to rear individually. Among them F1 female samples were observed with stereo, optical and electronic microscope. The brain ganglia of fourth-instar larvae were used for metaphase chromosome preparations. The air-dried slides were stained with Giemsa dye for further examination. Results The morphological differences were mainly on presence or absence of HP, pale spot on V2.1 and proboscis. These characters barely occurred in An. minimus A, but appeared in An. minimus C with a high proportion, especially the difference on pale spot on V2.1 was stable between An. minimus A and C. There was no difference to be found in the squama on the wing between An. minimus A and C examined by optical and electronic microscope. There were some differences between An. minimus A and C in the sex chromosome mitotic karyotypes on the ratio of short arm and long arm. Conclusion There are some stable differences between An. minimus A and C in morphology and chromosome mitotic karyotypes.

目的对微小按蚊A、C的形态和有丝分裂期染色体核型进行比较研究,观察并筛选具有鉴别意义的特征。方法将现场采集后经形态学鉴别初步认定为微小按蚊的样本带回实验室单管饲养,待其产卵后,经多重PCR方法鉴别为微小按蚊A或C。子代羽化后的成蚊标本使用解剖镜、光学显微镜和扫描电镜进行形态学观察和比较。取子一代四龄幼虫的脑神经节制作染色体核型,吉氏染色,显微镜下观察。结果微小按蚊A和C的形态学差异主要表现为是否存在膊白斑(HP)、V2.1白斑及喙腹面白斑,尤其是V2.1白斑和喙腹面白斑,微小按蚊A不出现或出现率极低,微小按蚊C则有很高的出现率,具有稳定差异。光学显微镜和扫描电镜下微小按蚊A、C翅前缘脉、V2.1及翅繸处鳞片及翅目上小钩均未见差异。微小按蚊A、C的常染色体着丝点指数和相对长度均无明显差异,性染色体均为近中着丝粒核型,但略有不同。结论我国微小按蚊A、C的形态和染色体核型均存在一定的差异。
