In addition, the polysaccharides were found to be able to inhibit the increase of gastric acid and to raise the content of PGE_2 in the gastric juice in rats treated with pentagastrin previously. But, no effects on the gastric acid and PGE_2 were found in rats treated with histamine and pilocarpin. However, when rats were treated with aspirin, PPA showed no effect on gastric acid and PGE_2. From the above results, it seems that the anti-ulcer action of PPA may be related to it's effect on the metabolism of PG. The polysaccharides may'bring about decrease of gastric acid secretion. Therefore, PPA could protect the gastric mucous membrane from injury.

硫酸基含量对多糖的生物活性起着至关重要的作用。运用硫酸钡比浊法测定梅花鹿鹿茸多糖(Pi-lose Antler Polysaccarides,PAPS)的硫酸基含量。采用可见分光光度法,最大吸收波长为360 nm,测得硫酸基含量5.8%。稳定性试验表明,硫酸基在60~120μg线性关系良好(r=0.997 19),同一样品在10~40 min内检测值比较稳定。回收率为97.25%。该方法比较准确、快速、灵敏度较高,适用于动物多糖硫酸基含量的测定。

Extraction Process of Pilose antler polysaccharides (PAPS) was optimized by response surface analysis and orthogonal method. The results showed that the optimum temperature,the reaction time,the pH value and the ethanol concentration of enzyme hydrolysis were 54℃,11h,8 and 70%,respectively. Extraction Process of Pilose antler polysaccharides (PAPS) was optimized by response surface analysis and orthogonal method. The results showed that the optimum temperature,the reaction time,the pH value and the ethanol concentration of enzyme hydrolysis were 54℃,11h,8 and 70%,respectively. Comparing the effects of deproteinization by Trichloroacetic acid and Tannin with deproteinization by enzyme and investigating the Enzyme-Sevag methods by orthogonal design,the method of enzyme-sevag was the best. When the ratio of polysaccharide and sevag solution was 1:1 and Trichloromethane / n-butanol (V/V) was 4.2:1,the deproteinization effect of sevag method was the best. The productivity of PAPS improved 48.35% as compared with traditional method.

通过响应面分析法与旋转正交回归法对鹿茸多糖的提取工艺进行优化,以期得到最佳工艺。应用响应面分析法对酶解温度、时间、pH值进行考察,得到最佳工艺参数为:温度54℃,时间11h,pH8;确定醇析最适浓度为70%;对三氯乙酸除蛋白法、单宁除蛋白法及酶-Sevag除蛋白法进行比较,并采用旋转正交回归法对酶-Sevag法进行分析,证明当多糖溶液:Sevag试剂=1:1,氯仿:正丁醇=4.2:1时,除蛋白效果最佳。应用最佳工艺后,多糖提取率比传统方法提高了48.35%。
