A rat model of minimal change nephrotic syndrome was induced by kidney artery infusion with the polycation protamine. A rat model of minimal change nephrotic syndrome was induced by kidney artery infusion with the polycation protamine. The changes of polyanion of the glomerular filtration barrier and the morphological changes of epithelial foot processes were studied using an Image Analyzer and stereological techniques. The results showed that filtration barrier polyanion decreased significantly, and the width and the volume fraction of epithelial foot processes increased. All of these pathological changes were caused by the destruction of the electrical charge of the kidney filtration barrier. These changes were also the main pathogenic bases of kidney filtration barrier permeability increase and heavy proteinuria.

We examined the effect of the superoxide ani-on[O2.] scavenger-superoxide dismutase (SOD) and non-oxygen free radicals scavenger-cyclo-sporin A (CSA) on ADR-induced nephrotic syndrome. The experiment found that the SOD can significantly reduce proteinuria, ameliorate serum biochemical index, and also attenuate renal injury. In particular, SOD also significantly suppress the serum and renal cortices malondialdehyde (MDA) concentration. Treatment with CSA was ineffective. The results suggest that the associated glomerular lesion in the ADR model is mediated by oxygen free radicals and lipid peroxides

The glomeru-lar alterations included swelling and fusionof foot processes and narrowing or loss offiltration slits. Electron-dense particles for-med by binding of prota mine to polyani-ons were seen primarily in the lamina raraexterna of glomerular basement membraneand also along the lamina rara interna andon the foot p rocess surface, which showedthe distribution feature of glomerularfilter. These results suggest that the ultra-structure alterations of glomeruli mayresult from the neutralization of polyanionsbyperfusion with protamine.

作者用多阳离子化合物鱼精蛋白灌注大鼠肾脏,引起极似人类微小病变型肾病综合征病理改变.结合电镜观察,用图像分析仪及体视学原理分别对该模型肾组织胶体铁染色切片和电镜照片上足突平均厚度及足突占毛细血管的体积分数进行形态计量学研究.结果表明:用药组肾小球多阴离子明显减少;足突肿胀、融合、滤过裂隙变窄甚至消失。鱼精蛋白与多阴离子结合形成的电子致密颗粒,见于足突表面及基底膜内、外疏层,并以基底膜外疏层为著。显示肾小球多阴离子的分布特点.提示肾小球超微结构的改变可能系多阴离子被鱼精蛋白中和所致.
