In this article, we investigated the effect of PHA and radiation, seperately or in combination, on the expression of LDH isoenzymes on rat thymus and lympho node cells. We found that the PHA of apropriate dosage was unable to raise the LDHS of lymphocytes nor the proportion of total M-type enzymes -After thfe irradiation with 250 rads from a 60Co source, lymphocytes showed a prominent decrease of LDH1 and an increase of LDH5. In this article, we investigated the effect of PHA and radiation, seperately or in combination, on the expression of LDH isoenzymes on rat thymus and lympho node cells. We found that the PHA of apropriate dosage was unable to raise the LDHS of lymphocytes nor the proportion of total M-type enzymes -After thfe irradiation with 250 rads from a 60Co source, lymphocytes showed a prominent decrease of LDH1 and an increase of LDH5. The differance waa very significant. The subgroup analysis showed that the; main effect of radiation was to depress the proportion of H type enzyme. The PHA in large dosage could obviously prevent the decrease in lymphocytes irradiated, though not change its LDH isoenzymes composition and spectrum distribution. It is suggested that the biological effect of radiation on lymphocytes might be affected by the functionary state of'target cells. On the other hand, our works may be valuable for further studies on the relation between the extrinsic stimuli and the functionary state of immunopotential cells.

本文把PHA和电离辐射两种因子,在单独和合并应用的条件下,观察了它们对大鼠胸腺和淋巴结细胞LDH同功酶表达所带来的影响。发现PHA在适宜剂量下,使T细胞LDH_5比例增高。但高剂量PHA对淋巴细胞的LDH_5或总的M型酶的比例均无明显影响。在经250rad~(50)Coγ射线照射后,淋巴细胞表现为LDH_1下降和LDH_5增高,但以LDH_1的下降最为突出,差别非常显著。从亚基分析来看,电离辐射主要使H型酶的比例下降。高剂量PHA虽未见改变正常淋巴细胞的LDH同功酶的组成与谱带分布,但具有明显地防止辐射所引起的LDH_1或H型酶显著下降和LDH_5或M型酶增高的作用。它提示辐射对淋巴细胞的生物效应,受到细胞所处功能状态的影响。这对进一步研究外界刺激的作用与免疫潜能细胞功能状态之间的关系,也有一定参考价值。

The experiments on the parent strain show-ed that ammonia could repress nitrogenase ex-pression completely, whereas glutamine at aconcentration of 10 mmol/L had no repressiveeffect (Fig. 3). In the presence of MSX, am-monia had no repressive effect on nitrogenasesynthesis (Fig. 4), but the effect of glutamine wasnot changed. When MSF, an inhibitor of GOGAT.was added to the medium, the expression ofnitrogenase in the parent strain was delayed(Fig. 5), and the addition of glutamine enhancedtha delaying effect. From the above results,the regulatory mechanism of nitrogenase syn-thesis and the role of GS-GOGAT pathway init were discussed.

浑球红假单胞菌的谷氨酸合成酶突变株的培养物缺乏固氮活性,在加入谷氨酰胺合成酶抑制剂MSX后,固氮酶得以表达。MSX加入后对突变株的氮源多效缺陷无恢复作用,说明氮源多效缺陷与固氮酶不合成分属遗传和生理两种水平的改变。当野生型菌株的培养物中加入MSF后,固氮酶表达延迟,谷氨酰胺的加入加剧这一延迟效应。
