Objective: Several kinds of chicken egg albumin (CEA) gradients were used to enrich the human sperm with good routine index. The enriching efficiency of each column was compared. Methods: 30 semen samples from normal healthy volunteers were used for this experiment. Sperms were treated sequentially with 7.5% single column single layer (DC), 7.5% and 17.5% single column two layer (SC) and 10%, 15.5% and 17.5% two column three layer (TC) through preselecting (FD) in 7.5% CEA gradienls: X and Y bearing sperm were detected by the quinacrine 2HCL fluorecent staining method. Results: Post treatment with CEA column. The sperm motility, percentage of progressively motile sperm increased remarkably ( P <0.05). The percentage of abnormal sperm decreased ( P <0.05). 34 79% recovery of sperm in FD group was significantly higher than that in DC (below 10%) and that of SC and TC (below 5%)( P <0.05). The positive ratio of Y body in DC, SC, and TC increased remarkably ( P <0.05). Conclusion: CEA could be used in selection of good quality sperm. The sperm from FD were more favorable than those from DC, SC and TC. The high positive ratio of Y body in the later three groups suggested that these kinds of CEA gradients could be used to enrich Y bearing sperms. The value of CEA to prevent X linked genetic disease by enriching Y sperm remained to be studied futher.

比较不同鸡蛋白蛋白( C E A)柱对 30 份正常生育力精液进行精子优选的效果。分别以 75%单层 一步法( D C)、75% 和125% 双层一步法( S C)及 75% 预 选( F D)再经 10% 、155% 和 175% 三层二步法( T C) C E A 柱进行优选。比较优选前后精子活动率、前向运动精子百分率和畸形率等常规指标及优选后各组精子回收率,并以盐酸喹啉染色比较优选前后精子 y 小体阳性率。结果:优选后各组精子常规指标显著提高,以 F D 组精子回收率最高(3479% ),高于上游法进行优选的回收率,与用人血清白蛋白进行优选的效果相当,可作为临床精子优选的有效手段。 D C、 S C 和 T C 各组回收率在 2% ~9% 之间,显著低于 F D 组,但 y 小体阳性 率均较优选前显著提 高, 在通过 X、 Y 精子 分离以预防 X 连锁遗 传病上的可能用途有待进一步研究。

Objective: To compare the relationship between the surviving time of the sperm cultured in vitro in oligoasthenoteratozoospermia and the pregnancy rate of IUI.Methods: Sixty patients of male factor infertility (oligoasthenoteratozoospermia) were included in this study. The sperm prepared by swin - up was divided into two parts. One was used to do IUI and the other one was used to determine the surviving time in vitro.Results: Male factor infertility was treated with IUI in 60 patients for 137 cycles. Nineteen patientsbecame pregnant with pregnancy rate (PR) of 31. 7% (19/60). Pregnancy rate per cycle was 13. 8% (19/ 137). There were 5 cases treated with IUI but no one was pregnant when the sperm surviving time (SST) was 9h; Ten cases were treated with IUI and 2 cases were pregnant with SST = 12h; Sixteen cases were treated with IUI and 4 cases were pregnant with SST= 24h; Twenty nine cases were treated with IUI and 13 cases were pregnant with SST>36h.Conclusions; The surviving time of the sperm cultured in vitro in patients of oligoasthenoteratozoospermia is of very great value in predicting the success of IUI. The pregnancy rate of IUI was significantly increased whn the SST ws longer than 36 hours.

为比较研究少弱畸形精子优选后体外生存时间与宫腔内人工授精(IUI)妊娠率的关系。选择因男方患少弱畸形精子症致不孕者。以改良F10培养液为精子培养液,采用上泳分离法体外处理精子后,置36.5℃二氧化碳培养箱中,分别于0,9,12,24,36,48h观察其精子活率和活力,活率≥50%者为阳性,活率<50%者为阴性。结果以生存时间(SST)表示。宫腔内人工授精按常规操作进行。共治疗60例,137个周期,妊娠19例,总妊娠率为31.7%(19/60),周期妊娠率为13.8%(19/137)。其中 SST 9h阳性5例,无一例妊娠,12h阳性10例,2例妊娠(20%),24h阳性16例,4例妊娠(25%),36h阳性29例,13例妊娠(44.8%),48h精子生存试验结果显示阴性,优选后精子体外生存时间对预测IUI妊娠率有重要临床价值,即SST为36h,人工授精妊娠率明显增高。

Objective To study and compare swim-up and Wang tube techniques in sperm separation. Methods 57 samples of semen were treated by two methods. Several characteristics of semen including sperm count, motility, morphology, acrosin morphology and normal chromatin were analyzed. Results There was significant difference between pre-treatment and post-treatment in both methods in terms of sperm motility and morphology. There was also significant difference on acrosin morphology and normal chromatin chromatin in Wang tube method, but not in swim-up method. Conclusion Both Wang tube and swim-up methods improve sperm quality, but Wang tube is superior to swim-up, especially in recovery rate of arcosin morphology and normal-chromatin sperm. It is a promising method in sperm separation for use in insemination and in vitro fertilization to improve birth health quality.

目的 探讨王氏管精子优选法在精子优选的应用 ,并与传统的精子上游法进行比较。方法 随机选择 5 7份精液 ,采用两种方法进行配对处理 ,比较两种方法分离前后、两者间精子活动率、形态以及精子顶体形态和精子染色质等参数的变化。结果 两种方法处理后的精子活动率、精子形态差异有显著性 (P <0 0 1) ,王氏管法处理后精子顶体完整率、正常染色质率亦显示出显著的差异 (P <0 0 1)。结论 王氏管精子优选法和精子上游法在精子处理中获得较好的精子质量 ,而王氏管精子优选法则获得更优质的精子 ,且具有方便操作实用的特点
