In the present paper some factors affecting the superinduction of interferon in diploid cells were studied. The results showed that the titer of interferon superinduced in fetal skin muscle cell cultures was significantly higher than those in fetal human lung cell cultures. Cells treated with Astragalus membranaceus produced more interferon. Poly I:C and cycloheximide could be present simulstaneouly in the cell cultures during the whole course of superinduction. In the present paper some factors affecting the superinduction of interferon in diploid cells were studied. The results showed that the titer of interferon superinduced in fetal skin muscle cell cultures was significantly higher than those in fetal human lung cell cultures. Cells treated with Astragalus membranaceus produced more interferon. Poly I:C and cycloheximide could be present simulstaneouly in the cell cultures during the whole course of superinduction. However, the duration of treatment with inhibitors of nucleic acid synthesis ( mithramycin and actinomy-cin D ) should not exceed one hour before the termination of superinduction. The. behaviour of superinduction by mithramycin was similar to that by actinomycin D The optimal time for combined poly I : C and cycloheximide treatment was about 9 hours. Under our experimental conditions, the interferon yield in the superinduction system was increased by 170-248 fold as compared with similar system without superinduction. The present paper describes a procedure for the large scale production of fibroblast interferon.

本文在人二倍体细胞培养上就影响干扰素超诱导的某些因素进行了研究。结果表明:人肌皮二倍体细胞株的干扰素超诱导能力高于人肺二倍体细胞株。黄芪处理细胞在超诱导系统中可以生产较高量的干扰素。聚I:C和环已亚胺能在超诱导的整个过程持续存在,但放线菌素D和光辉霉素则在超诱导终止前作用1小时最适宜。光辉霉素与放线菌素D有相似的干扰素超诱导作用。聚I:C与环已亚胺的适宜作用时间为7~9小时。在适宜条件下,干扰素的产量在超诱导系统中比单诱导增加170~248倍。本研究可为大量生产纤维细胞干扰素及提取干扰素mRNA提供依据,并为阐明干扰素超诱导机理提供基础。

The metabolism of NaHSO_3-andrographolide in rats was studied by means of NaHSO_3(~(35)S)-andrographolide.

本文是用放射性同位素~(35)硫标记NaHSO_3-穿心莲内酯做动物实验,以阐明NaHSO_3(~(35)S)-穿心莲内酯在体内的代谢过程。动物实验是用NaHSO_3(~(35)S)-穿心莲内酯注入大白鼠。实验用的药物是用NaHSO_3(~(35)S)溶液用加成反应标记于穿心莲内酯的第12位碳上。标记率达92%。标记化合物的性能和浓度与治疗用商品注射针剂NaHSO_3-穿心莲内酯相同。大小便标本是注射后连续按一定间隔时间取出的,组织标本是按一定间隔时杀死的动物身上取出的。标本的放射性是用液体闪烁计数器测量的。标记化合物在大白鼠的体内代谢表明,NaHSO_3(~(35)S)-穿心莲内酯进入体内后,在中枢神经系统中主要选择性地蓄积于脊髓中,并且呈下行性蓄积,可因此导致肌紧张度减低和外周血管扩张,引起体温下降。在脏器中此药较明显地蓄积于十二指肠、直肠,这是临床采用此药治疗肠炎和菌痢的有力依据。实验也指出,此药在体内迅速吸收,迅速排泄,72小时内在尿粪中总排除量达86.2%,建议临床酌情增加剂量或次数。NaHSO_3(~(35)S)-穿心莲内酯大多在尿中主要以原形排出,从而考虑对泌尿系统感染可能有一定疗效。

According to the view point of drug metabolic regulation, the effects of six aromatic amines and amides on isoniazid acetylated metabolism were studied in rabbits. All of four aromatic amines inhibited acetylation of isoniazid, but amides did not. 4-Aminobenzoic acid can be considered as a combined-drug for increasing and prolonging the blood level of isoniazid. The effects of INH, PAS and INH plus PAS on excretion of vitamin B_6 were also studied.

根据药物代谢调节的观点,在家兔体内研究了六种芳香胺和酰胺类化合物对异烟肼乙酰化代谢的作用。四种芳香胺都具有抑制乙酰化代谢的作用,而酰胺类没有这种作用,并提出对氨基苯甲酸可考虑作为合并药物以提高异烟肼的血水平和持续时间。还研究了异烟肼、对氨基柳酸以及两者合用时对维生素B_6排泄的影响。
