In the androgenetic diploid tadpoles(nucleo-cytoplasmic hybrids)obtained,the appearance of pigments in the eyesand melanophores in the skin occurredat the same stages as in wild-type,butmuch earlier than in the albino mutant.Plenty of melanophores appeared in theskin,especially e.g.,on the dorsalsurface,on the sides of abdomen,as wellas on the tail.These are characteristicsof the wild-type and have never beenseen in the a~p mutant.Therefore the de-velopment of pigmentation in hybridsare believed to be contributed by Xenopuscytoplasm.During metamorphosis,themelanophores began to depigment. Two of the androgenetic diploid toad-lets have grown to adults and have beenused for the following experiment.Eggsof a~p mutant were fertilized with spermsof the androgenetic diploid.The fertilizedeggs developed very well.Of the 524eggs,470 embryos hatched and most ofthem metamorphosed into healthy toadlets.It is clear that the anurna amphibia tad-poles with only paternal genetic materialcan develop into adults and produce nor-mal gametes.

Androgenetic diploid clones of Paramisgurnus dabryanus were induced by heat shock. The method was as follows: the eggs of Misgurnus anguillicaudatus were immersed in synthetic ovarian fluid and irradiated by UV with the dosages of 210mJ/cm~2 to inactivate genomes of eggs, then the eggs were fertilized with the sperm of P. dabryanus. Each group of eggs was applied by heat shock at 39℃ for 2min to inhibit the first mitotic division of haploid androgenesis 15min after fertilization, at intervals of 2min each. The results show that two of eggs with high yields of androgenetic diploid occurred in 15~19min and 27~29min after fertilization, the highest yield was 61.1%. According to chromosome and morphology of P. dabryanus, their chromosome numbers (2n = 48) and morphology were all-paternal inheritance.

The eggs of zebrafish (Danio rerio)were irradiated by shortwave ultraviolet (UV) of 120 mJ/cm~2 for inactivation of maternal nuclei. The dapple skin was used as genetic marker, and heat shock was used to double genome. It is validated that leopard dapple skin is homozygotic recessive mutation and is applied as an androgenetic genetic marker. The androgenetic diploids occurred within 2 min, heat shock at (41.5±0.5)°C at 11 min and 23 min after fertilization, respectively.

为探索人工诱导斑马鱼雄核发育二倍体克隆鱼的途径和方法,运用紫外线照射卵子使其遗传物质失活,利用斑马鱼体表花纹形态作为遗传标记,使用热休克技术阻止受精卵的第一次卵裂加倍染色体。试验结果表明总辐射剂量为120mJ cm2紫外线照射斑马鱼卵可以使雌核遗传物质完全失活;体表花纹为豹斑花纹被证实是由纯合隐性基因控制的性状,可以作为雄核发育遗传标记;遗传失活卵受精后经2min,(41 5±0 5)℃的热休克后出现两个染色体加倍的高峰期,分别为受精后的11min和23min,且后一时期效果优于前一时期,最高二倍体诱导率达46 15%;经遗传分析,共获得正常存活的雄核发育二倍体克隆鱼19尾,说明经过优化的紫外线结合热休克等染色体操作技术,可以成为诱导雄核发育二倍体克隆鱼的有效手段。
