Five new 4-substituted-2,2,6,6-tetramethyl-piperidine-1-oxyls 6(a~e)were synthesized.Their structures were ascertained by elemental,IR and ESR spectraanalyses.Experimental results showed that the compounds 6(a~c) possessed inhibitingactivity on L7712-leukemia cell.

本文报道了五种新型的4-取代硫脲基-2,2,6,6-四甲基哌啶氮氧自由基化合物6(a~e)。并经元素分析、红外光谱及电子自旋共振波谱确证了它们的结构。药理实验表明,化合物6(a~c)对 L7712白血病细胞具有一定的抑制作用。

Organic compounds with low molecular weight and humic acid in water sample were separated and concentrated by using resin GDX-102. The incorporations of [3H] thymidine, [3H] uridine and [3H] leucine into DNA, RNA, and protein respectively in leukemia 7712 cells were inhibited by the comprex of 2. 2, 6, 6 - tetramethy 1 - 4 - isothiocyano-piperidine-1- oxyl and 2, 2, 5, 5 - tetramethy 1 - 4-isocyano-pyrrolion - ocyl with praseodymium dicitrite 50 μg/ml for 3 - 24 hours. The degree of inhibition increased in proportion to the incubation time. After removing the complex from the medium entirely, the rate of DNA synthesis increased with time over 1-7 hours. This result indicated that the inhibition mechanism was likely metabolic lock agent for DNA synthesis.

2,2,6,6—四甲基哌啶氮氧自由基等自由基化合物,均比其还原体化合物对癌细胞DNA合成的抑制率高。四甲基哌啶氮氧自由基和四甲基吡咯啉氮氧自由基对L7712细胞DNA,RNA和蛋白质合成的抑制作用,在3—24小时内均随作用时间延长而增高,它们对癌细胞DNA合成的抑制作用机理,似属干扰代谢型。
