Sequencing analysis of lipoprotein lipase ( LPL ) gene exons 1 9 was made in 11 cases of Chinese patients with severe endogenous hypertriglyceridemia (serum TG >7.68 mmol/L) by the dideoxy chain termination method using Sequenase PCR Product Sequencing Kit. This was a conservative mutation with an allele frequency of 11.4% in the 74 of the Chinese controls.Two others Ala 261 Thr (G 1037 A) in exon 6 and Ser 447 Ter (C 1595 G) in exon 9, which were also found in Caucasians and have been reported to have disease associations with dyslipidaemia,showed at frequencies of 0% and 10.7% in the Chinese controls respectively.However, it is the first time that these three genetic variants have been reported in Chinese subjects.The results show that the gene variation of LPL may not be associated with endogenous hypertriglyceridemia in Chinese population.

研究表明:氧化LDL(Ox-LDL)可抑制LPS诱导的巨噬细胞NO释放,而正常LDL和乙酰化LDL则无抑制作用。用谷胱甘肽过氧化物酶模拟物ebselen清除Ox-LDL上的脂氢过氧化物对其抑制作用没有影响。Ox-LDL的蛋白组分对NO释放也没有影响,而脂质组分则有抑制作用。LDL脂质的主要成分亚油酸和卵磷脂无论单独还是共同氧化后对NO释放都没有影响,而亚油酸和胆固醇一起氧化后则对NO的释放有很强的抑制作用,胆固醇单独氧化后也有一定的抑制作用,提示氧化胆固醇可能是Ox-LDL对NO释放起抑制作用的主要因素。

AIM: To investigate the effects of new antioxidant, seleno Glutathione peroxidase PZ51 on endothelial nitric oxide synthase (eNOS) protein expression of coronary artery in Stroke Prone Spontaneously Hypertensive rat (SHRsp) during the chronic process of hypertension. METHODS: 22 SHRsp of 8 week old which were randomly divided into PZ51 group and control group were administered by gavage for six weeks. Heart weight ratio, heart structure, the level of nitric oxide (NO) of myocardium and eNOS protein of coronary artery was examined. RESULTS: Compared with the control group, PZ51 significantly increased NO of myocardium homogenate ( 1.18 ± 0.25 vs 0.72 ± 0.18 μmol·mg -1 prot, P< 0.05 ) and eNOS protein expression( 7.45 ± 2.10 vs 2.77 ± 2.07 ,P< 0.05 )of coronary artery. CONCLUSION: After 6 weeks administration of PZ51, the eNOS protein expression of coronary artery and NO level of myocardium in SHRsp significantly increased.

目的 :研究新型抗氧化剂含硒谷胱甘肽过氧化物酶模拟物PZ5 1对卒中易感型自发性高血压大鼠 (SHRsp)高血压发展的慢性过程中冠状动脉内皮型一氧化氮合成酶 (eNOS)表达的影响。方法 :2 2只8周龄SHRsp随机分为PZ5 1组和对照组 ,灌胃治疗6周。测量心脏重量比值、心脏结构 ;采用分光光度计测心肌组织匀浆一氧化氮 (NO)的浓度 ;免疫组化检测冠状动脉eNOS蛋白表达。结果 :PZ5 1组较对照组心肌组织匀浆NO浓度显著升高 (1.18± 0 .2 5vs 0 .72± 0 .18μmol·mg-1prot ,P <0 .0 5 )、冠状动脉内皮eNOS蛋白表达显著增加 (7.4 5± 2 .10vs 2 .77± 2 .0 7,P <0 .0 5 )。结论 :PZ5 1治疗 6周显著增加了SHRsp冠状动脉eNOS蛋白表达及心肌组织NO的水平。
