The reactivity of the various components of the Golgi apparatus of rat spermatids for three phosphatase activities (nicotinamide adenine dinucleotide phosphatase, NADPase; thiamine pyrophosphatase, TPPase; cytidine monophosphatase, CMPase) and the incorporation of 3~H-fucose by the spermatids were analysed at the four phases of spermiogenesis. The reactivity of the various components of the Golgi apparatus of rat spermatids for three phosphatase activities (nicotinamide adenine dinucleotide phosphatase, NADPase; thiamine pyrophosphatase, TPPase; cytidine monophosphatase, CMPase) and the incorporation of 3~H-fucose by the spermatids were analysed at the four phases of spermiogenesis. The results indicate (1) that the Golgi apparatus of spermatids undergoes a series of morphological and cytochemical changes during spermiogenesis; (2) that the Golgi apparatus of spermatids, although it loses its CMPase positive GERL element in acrosome phase, retains evidence of functional capacity until it degenerates in maturation phase; (3) that in early spermatids the various saccular components of the Golgi are specialized with respect to enzymatic activities; and (4) that each Golgi region may contribute in a coordinated fashion to the formation of the acrosome system and multivesicular bodies.

Because of the obscurity in the originator of L7912, an initial study on this problem was carried out through comparing L7912 with L7712 in the karyotype, ultrastructure and cytochemistry.

After treatment with RA or DMSO, HL-60 cells underwent a series of morphological changes in nucleus and cytoplasm, and functional changes in phagocytosis and NBT reduction. MPO reaction revealed that MPO localized in endoplasmic reticulum. nuclear envelope lumen and azurophil granules in the early differentiation stage and appeared only in azurophil granules in the later stage. Both ultrastrueture and cytochemical reaction showed that the HL-60 cells treated by RA or DMSO had gradually differentiated into mature granuloeytes, but some of those treated by DMSO differentiated into macrophage-like cells.

用电镜及超微结构细胞化学方法,观察并比较了RA及DMSO诱导HL-60细胞分化的演变过程,两种诱导剂均可使HL-60细胞分化为有吞噬功能的粒细胞,但DMSO处理组尚出现少量分化为单核细胞。超微结构和细胞化学的结果说明RA诱导HL-60细胞分化有一个清晰的成熟演变过程。在诱导剂处理四天中,从最初表现为核变形至分裂、胞浆A颗粒由疏松变为致密以及S颗粒和吞噬活动出现;MPO反应从开始时出现于内质网、核膜腔、A颗粒,演变为最终只存在于A颗粒。DMSO处理细胞在第二、三天间有一个诱导分化的转折点。对上述结果进行了初步讨论、
