    Effect of glutamate on hydroxyl radical in rat corpus striatum has been studied.Based on the pashpull microperfusion system and HPLC-UVdetection,we found that the concentration of 2,5-DHBA induced by salicylate capturing hydroxyl radical in the microperfusate was significantly increased during 15 ̄30 mn of perfusing glutamate(0.1mmol·L-1)to corpus striatum.The increasing concertration of 2,5-DHBA was partly reduced by perfusing glutamate and ketamin (0.6mg·ml-1)simultaneously.The results suggest that increasing...

    采用脑区微灌流系统和水杨酸为捕捉剂的HPLC羟自由基检测法,观察到大鼠纹状体灌流0.1mmol/L谷氨酸15~30min内,灌流液中羟自由基和水杨酸反应生成的衍生物2,5-DHBA的浓度为310.4±171.8ng/ml(x±s,n=5)较谷氨酸灌流前的84.8±37.0ng/ml(x±s,n=5)含量显著增高(P<0.01).而给予氯胺酮0.6mg/ml加灌组,谷氨酸灌流15~30min内的2,5-DHBA浓度为165.4±88.1ng/ml(x±s,n=5)。结果提示纹状体细胞外谷氨酸含量增加能使局部羟自由基生成增加且能被氯胺酮部分抑制。

    Aim:In order to investigate how hyperkalemia’protects myocardium agaenst is chemia and reperfusion(I-R)injury。Materials and Methods electron-microscopy,phosphate-pyroantimonate(PPA)calcium localizaton techniques and enzyme testing methods were used to investigate the changes in the isolated rat hearts。Results are as following:13 mmol/L potassium protects the activity of superoxide dismutase(SOD);reduces the content of malondialdehyde(MDA);decreases the release of lactate dehydrogenase(LDH);preserves membrane's ability to combine calcium;prevents Ca++ accumulatein in mitochondria(Mi)and alleviates cell ultrastructural injury。Conclusion:The results show that l3mmol/L potassium protects the myocardial I-R injury by depressing intracellular calcium overload and membrane lipid peroxidation.

    为了解高钾抗心肌缺血再灌注(I-R)损伤的途径。用离体大鼠心脏行冠脉结扎,松扎后作电镜观察和磷酸盐-焦锑酸盐(PPA法)细胞化学钙定位及抗氧自由基检测。结果:13mmo1/L高钾保护I-R心肌超氧化物歧化酶(SOD)活力,抑制丙二醛(MDA)生成,减少心肌酶的漏出,保存肌膜结合钙的能力,防止钙在线粒体的积聚,减轻心肌超微结构的破坏。实验提示13mmol/L钾能抑制胞内钙超负荷和膜脂质过氧化达到抗I-R损伤作用。

    To study the free radicals biology and free radicals medicine, methodolgy is of critical importance. Many methods have been used to measure free radicals in biological systems, but everyone has some advantage and also some disadvantage in different experiments. Recently people often use some advantage methods, such as electron spin resonance(ESR), lipid peroxidation, free radicals cleaning enzymes and so on.

    检测生物体系中自由基的方法是一种开展自由基生物学和自由基医学研究的重要实验手段.生物体中自由基的检测方法有很多,但各种检测方法在不同的实验中都有各自的优缺点.近年来应用较多的实验方法是电子自旋共振法(ESR法)、脂质过氧化法、自由基清除酶法等.
