Objective To explore the mechanism of obesity and chronic anovulation in androgen sterilized rat at age of 9 days (9d ASR) and the effect of herbs of tonifying Kidney yin. Methods 9d ASR model was established by injecting testosterone propionate subcutaneously to SD female rat at age of 9 days. Double immunofluorescent staining was carried out on sections through arcuate nucleus (ARC) to determine whether neuropeptide Y (NPY) containing neurons expressed estrogen receptors (ER). The immunoreactive amounts of ER, NPY neurons and gonadotropin releasing hormone (GnRH) fibers of hypothalamic ARC median eminence (ME) in 9d ASR were studied with immunohistochemistry. Serum estrogen (E 2), testosterone (T), leptin, follicular stimulating hormone (FSH) and luteinizing hormone (LH) were measured with radioimmunoassay. All parameters were reevaluated after administration of herbs to 9d ASR. Results Estrogen receptors were predominantly distributed in the cytoplasm of NPY containing neurons as well as in the nucleus and axons of NPY synthesizing neurons. Compared with normal group, ASRs were characterized by high metabolism rate, obesity, increase in hypothalamic NPY expression (P<0.01), decrease in numbers of ER and GnRH secretion (P<0.01). Levels of serum E 2, T and leptin were significantly higher and FSH and LH levels significantly lower in 9d ASRthaninnormalrats (P<0.01). Significantly negative correlation between NPY and ER or GnRH (r=0.7104, P<0.01; r=-0.8802, P<0.01, respectively) were observed, andsowerethecorrelationsbetweenleptinandFSHor LH (r=-0.7517, P<0.01; r=-0.8444, P<0.01, respectively). Serum leptin showed positive correlation with body weight in 9d ASR (r=0.8977, P<0.01). These dramaticneuroendocrine metabolicchanges becamenormal after feeding 9d ASR with herbs. Conclusions Obesity and chronic anovulation coexist in 9d ASR. Elevated E 2 levels may down regulate the hypothalamic ER oriented NPY neurons, followed by the release of NPY and suppression of GnRH. Effect of the herbs may be involved in reducing body weight and inducing ovulation.

The seminal fluid was analyzed and FSH (follicle stimulating hormone), LH (luteinizing hormone), T (testosterone), PRL (prolactin) and E2 (estradiol 2) were detected before and after treatment. The total effective rate was 92.1%. The mobile rate, mobility of sperm, and sperm count were all improved. The contents of FSH, LH and T were enhanced obviously.[Conclusion] JSW can improve the sperm quality and has a good effect on male infertility with kidney yang deficiency.

[目的]探索男性不育症中药治疗效果。[方法]选择符合肾阳虚型、少、弱精症诊断标准的男性不育症患者38例 ,予以金匮肾气丸口服治疗。治疗前后作精液分析 ,并检测促卵泡激素 (FSH)、黄体生成素 (LH)、睾丸酮 (T)、泌乳素 (PRL)和雌激素 (E2)。[结果]38例患者显效29例 ,有效6例 ,无效3例 ,总有效率92 1 %。患者精子活率、精子活力、精子总数均明显提高。FSH、LH和T均有明显上升。[结论]金匮肾气丸能改善精子质量 ,有效治疗肾阳虚型男性不育症。

Objective: To investigate the effects of electroacupuncture (EA) of "Guanyuan" (CV 4) and "Sanyinjiao"(SP 6) on neuroendocrine function in perimenopause model rats.Methods:Rat perimenopause model was made by removing the ovarium. 40 SD female rats were randomly divided into control group, EA group, model group and estradiol group, with 10 cases in each group. Serum estrin (E)2, follicle stimulating hormone (FSH), luteotropic hormone (LH) and β-EP of the hypothalamus were determined with radioimmunoassay before and after EA of SP-6 and CV-4 (2~100 Hz, 2.5 V, 20 min/time, once daily, 10 days/therapeutic course). In estradiol group, muscular injection of estradiol (0.05 μg/10 g) was given to the rats, once every other day. Results:In comparison with control group, serum E 2 and hypothalamic β-EP of model group decreased significantly (P<0.05), serum LH and FSH of model group increased markedly (P<0.05). Compared with model group, serum E 2 and hypothalamic β-EP of EA group and estradiol group increased apparently (P<0.05), while LH and FSH of these two groups lowered remarkably (P<0.05). Findings indicated that EA treatment could significantly raise serum E 2 and hypothalamic β-EP, and obviously lower serum LH and FSH levels. Conclusion:Electroacupuncture of "Guanyuan" (CV 4) and "Sanyinjiao"(SP 6) can regulate neuroendocrine function in ovariectomized rats, suggesting EA may play a role in improving menopause syndrome.

目的 :研究电针“关元”、“三阴交”对围绝经期模型大鼠神经内分泌的调整作用。方法 :复制大鼠去卵巢模型 ,测定电针“关元”、“三阴交”对模型大鼠血清雌激素 (E2 )、促卵泡激素 (FSH)、黄体生成素 (LH)和下丘脑 β EP含量的影响。结果 :模型组较正常组血清E2 含量降低 ( 2 2 77±0 1 3 5ng/L) ,FSH、LH含量升高 ( 7 0 6 2± 1 3 56mIU/mL ,3 473± 0 3 55mIU/mL) ,下丘脑 β EP含量显著降低 ( 1 6 751± 1 484pg/mg) ;电针组较模型组血清E2 含量显著提高 ( 3 44 9± 0 1 53ng/L) ,FSH、LH含量显著降低 ( 5 71 4± 0 3 50mIU/mL ,2 6 3 8± 0 1 98mIU/mL) ,下丘脑 β EP含量显著升高 ( 2 2 1 2 2± 0 82 8pg/mg组织 )。结论 :电针“关元”、“三阴交”治疗围绝经期综合征通过调节生殖内分泌和调整自主神经功能紊乱发挥作用
