在摇瓶试验基础上,采用里氏木霉(Trichoderma reesei)HC-415菌株进行5L自控罐产纤维素酶深层发酵试验。在通气量为 0.2—0.6vvm、搅拌速度为 400r/min、发酵液pH控制在5.8—6.1的条件下,发酵液的羧甲基纤维素(CMC)酶酶活最高为325.0mg糖/ml,滤纸糖酶(FPA)酶活最高达17.9mg糖/ml。发酵周期为108h。所得冻干纤维素酶粉CMC酶活最高3111IU/g,FPA最高135IU/g ,对发酵液得率平均6.7g/L。酶活总收率CMC酶活平均78.2%,FPA酶活平均73.5%。

The Cellulase enzyme was produced on a spent grain medium with solidstate fermentation process by Trichoderma reesei. The results indicated that the filter paper activity(FPA) attained 357U/g after 144 hours of fermentation by turning koji in 48h. Adopting liquid inoculum that was cultured in medium contain 3% wheat bran at pH6.5, FPA activity is 178U/g.

利用里氏木霉(Trichodermaresei),以啤酒厂的废糟为原料,添加适量麸皮和稻草粉为培养基进行固态发酵,采用固体种曲混合接种,在48h翻曲,经144h发酵后FPA酶活达到357U/g。以补加3%麸皮的酒糟水为培养基,调起始pH6.5培养92h的液体种子接种,FPA酶活为178U/g。

以麦糟为主要原料 ,利用里氏木霉 (TrichodermareeseiRutC 30 )固态发酵生产纤维素酶 ,在浅盘发酵中 ,通过对发酶过程品温和曲料pH的优化控制 ,得到了优化后的工艺条件。在发酵前期品温控制在 2 7~ 30℃ ,后期最高不超过 35℃ ,最高酶活达到 611U/g曲。当曲料pH第一次回升至 4 .3~ 4 .5时 ,用稀盐酸进行喷酸控制。采用优化后的pH和品温控制工艺 ,FPA酶活达到 50 8U/g曲 ,相当于 2 12 0U/g纤维素
