Based on the human, rat and rabbit SRY homologous sequence, a pair of primers was designed to amplify cattle Sry fragment with PCR. With our new male-specific cattle Sry sequence, a pair of primers specific to cattle Sry was developed to sexing preimplanted embryo cells and to detect bovine fetal Sry sequence from maternal peripheral blood. The results showed that bovine Y-specific Sry sequence can be detected directly from preimplanted embryo cells for sex manipulation and from maternal blood for non-invasive...

Objective:To establish the method for prenatal sex diagnosis of fetus carrying sex-linked genetic disorder.Method:Adopted PCR techni-que with x-y homologous gene and SRY gene (DYS388),analyzed 22 cases amnioticc fluid of pregnant women.Result:Male revealed three bands:106bp,112bp and 150bp and female only showed one band which was 106bp.conctusion:The results were accurate and reliable.It will be suitable for clinical prenatal diagnosis of sex-linked genetic disorder by using PCR with x-y homologous primers....

目的 :建立一种对性连锁遗传病胎儿进行产前诊断的方法。方法 :采用X -Y同源序列和SRY基因的两对引物 ,进行聚合酶链反应复合扩增。结果 :对 2 0例中孕羊水进行了性别签定 ,男性均显示 10 6bp ,112bp和 15 0bp3条扩增带 ,女性只有 10 6bp 1条扩增带 ,性别签定准确可靠。结论 :应用X -Y同源引物和SRY基因引物复合扩增 ,可用于临床上性连锁遗传病的产前性别诊断 ,方法可靠准确。
