We used coated SPC-A-1,the cell lineof human lung adenocarcinoma with mouseanti-normal human lung tissue antibodies asimmunogen to immunize BALB/C mice.Sple-ens taken from 3 days after last injectionwere fused with mouse myelomas NS-1 andSP 2/O.Seven monoclonal antibodies chosenrandomly from 75 hybridomas after cloning3—7 times were screened with a variety ofnormal human adult tissues,tumor and embr-yonic tissues with ELISA,immunofluorescenceand ABC staining. 1.To answer the question that whetherthe SPC-A-1 adenocarcinoma cells and othertumor cells share the common antigens orcross reactive antigens with normal lung tis-sue,we incubated the anti-normal lung tissueantibodies with three established tumor celllines and detected by immunofluorescence me-thod.Results in Table 1 indicate that threedifferent tumor cell lines react more or lesswith anti-normal lung Ab.ranging from 28.0% to 47.8% and that not all the cells inthe population are positive,indicating thatthe cells in SPC-A-1 cell line are heteroge-neous and that "coating" the cells showingpositive reactions with anti-normal lung tissueantibody could diminish the antigenicity ofnormal antigens. 2.The cross reaction of anti-"coated"SPC-A-l antibody with some tumor cells.Results in Table 2 show non-absorbed antiSPC-A-1 antiserum can react with both ade-nocarcinoma cell lines,SPC-A-1,SPC-A-2. However,when this antiserum absorbed withSPC-A-1,all the reaction with these threecell lines disappeared,when antiserum wasabsorbed with SPC-A-1 and SPC-A-2 cellsseparately it still retains some reaction withSPC-A-1 cells.These results indicated thatthere was presence of common antigenic deter-minants on both SPC-A-1 cells and SPC-A-2cells but SPC-A-1 had some other determina-nts which were absence on the SPC-A-2 cells.The fact that antiserum against the coatedSPC-A-1 cells did not react with SMC-1 cellsindicated that after coating,the normal lungantigenic presented on the tumor cells disap-peard or diminished their antigenicity in vivo. 肺腺癌细胞株SPC-A-1,经抗人正常肺抗血清复被后免疫小鼠,脾细胞和同系NS-1或SP 2/0融合,获得17株单克隆抗体。本文报道其建株过程及其中7株单克隆抗体的一些特征。LAC-122、LAC-163与LAC-210对人体肺腺癌和肺鳞癌的阳性反应率分别为75%、78.3%和75%,除LAC-122对一例胃癌细胞株有微弱交叉反应外,三株单抗对肺小细胞癌, 其它肿瘤组织及正常、胚胎组织均呈阴性。对利用抗体复被瘤细胞及肺癌相关抗原问题作了较详细的讨论.

A mouse lung adenocarcinoma(UL)induced by urethane was serially transplanted in the subcutaneous tissue of BALB/C nude mice. Light-and electron-microscopic studies comfirmed its origin of type Ⅱ alveolar epithelium. Certain features characterized this tumor model: (1) high take rate(100%), (2) rapid growth, (3) apparent invasiveness and high frequency of recurrence after excision of the primary implantation, (4) spontaneous lung metastases from the subcutaneous tumor and (5) marked elicitation of angiogenesis...

A mouse lung adenocarcinoma(UL)induced by urethane was serially transplanted in the subcutaneous tissue of BALB/C nude mice. Certain features characterized this tumor model: (1) high take rate(100%), (2) rapid growth, (3) apparent invasiveness and high frequency of recurrence after excision of the primary implantation, (4) spontaneous lung metastases from the subcutaneous tumor and (5) marked elicitation of angiogenesis and firbroblast proliferation in the adjacent host tissues. The above features render this animal model useful in studying tumor invasion and metastasis, and tumor angiogenesis as well.

我们用Urethane诱发的小鼠肺腺癌在裸小鼠皮下移植传代并研究了该肿瘤的生长及转移特性。此转移模型具有移植成功率高,生长迅速,血管血窦丰富,并有较高的自发肺转移倾向,浸润性强等特点。这对于研究肿瘤转移、浸润以及肿瘤血管生成具有一定意义。

The effect of PH on Plase C activities of Anip_(1211) and AGZY_(83-a) was studied. The optimal pH of Anip_(1211) and AGZY_(83-a) PIase was found to be 5.0, 6.0 respectively. AGZY_(83-a) PIase specific activity was 3.4 times of Anip 1211 at pH 5.0. Both enzyme activities exhibited in absence of Ca~(2+).

本文报道pH对不同转移表型肺腺癌细胞磷脂酰肌醇特异的磷脂酶c(PIase C)活性的影响。两条pH-酶曲线分别显示两个最适pH。在pH 5.0条件下,AGZY_(83)-a PIase C的比活力是Anip_(1211) PIase c的3.4倍。两者对Ca~(2+)的依赖性均较弱。
