For the purpose of taking the advantage of the superiorities in the fishing district where the aquatic resourses are rich and raw materials are fresh, the effect of removal of the protein from nucleoprotein of fish spermary (fish wastes in the aquatic processing industry) has been studied. Based on the modified SDS method, SDS-CHCl_3 method and phenol method, these studies compare the different effects of the removal of protein when varying the concentration of nucleoprotein and the composition and the dose of the protein denaturant etc. Through a good deal of experiments, the best technological parameters are determined and a new practical process suitable for the condition of the domestic commercial production is laid down. The analytical result of the product extracted by this technological process is that DNA content is more than 90%, the ratio between nitrogen and phosphorus varies from 1.62 to 1.80', the Extinction coefficient (one gram atomic phosphorus) ∈ (p) ranges from 6,500 to 7,500, the molecular weight (over 80 per cent of the total) is larger than 2.5×10~6 doltons. These results show that the spermaries from the first three species of fishes may be considered to be ideal materials. Some preliminary suggestions have been put forward about the test standard of the quality and analytical methods of the product (BR grade).

本文报告了利用渔区水产资源丰富和原料新鲜的优势,采用水产品加工的下脚料(鱼精巢),以改良SDS、SDS-CHCl_3和酚法为基础,通过改变核蛋白浓度、变性剂的构成与用量等方式研究了从鱼精核蛋白(粗)中除去蛋白质的效果,通过大量试验,确定了生产高聚DNA Na的最佳工艺参数与制定了适合国内生产条件的新工艺流程。本工艺制得的产品分析结果是。DNA含量大于90%,氮磷比为1.62—1.80,克原子磷消光系数ε(p)为6500~7500,分子量(80%以上)在2.5×10~6道尔顿以上。这些结果表明:鲐、兰园鲹、大黄鱼的精巢是生产高聚Na-DNA的理想原料。

对用 SDS法、尿素法、改良 SDS法、氯化苄和 CTAB法提取的大白菜基因组DNA进行了比较。结果表明 ,改良 SDS法提取的 DNA具有典型的天然 DNA分子的标准紫外吸收光谱特点 ,其 A2 6 0 /A2 80 为 1 .7~ 1 .9,A2 6 0 /A2 30 为 1 .8~ 2 .0 ,叶片的 DNA产量为 40 0μg/g。用该法提取的大白菜 DNA适于进行 RAPD分析。

Based on the analysis of several isolation methods for genomic DNA of Chinese chestnut,it was found that the improved SDS method was optimal for RAPD and other molecular biology researches.

通过对板栗基因组 DNA几种提取方法获得的 DNA质量分析 ,表明改良 SDS法是最为理想的提取方法 ,能满足 RAPD及其他分子生物学研究的需要
