目的 :ABO亚型在临床输血中经常造成错定血型及输血反应 ,因而研制抗A1试剂血清是解决ABO亚型鉴定的主要途径。方法 :采用血型血清学特异性抗原抗体吸收实验。结果 :经大量反复实验 ,研制出特异性强 ,效价符合要求的抗A1试剂血清。结论 :抗A1血清是鉴定A亚型的专用试剂血清 ,对临床鉴定ABO亚型及安全输血有实用意义。

Objective To establish a new flow cytometry-based method to identify the subgroups of ABO. Methods Human source anti-A(B)labeled with FITC(anti-A(B)-FITC) were specifically bound to human RBC. The fluorescence dot plot diagrams of each cells were obtained by FCM. After gating RBC, the diagrams were analyzed by using software-cell quest verl 2.2.Results The anti-A-FITC and anti-B-FITC of blood group A RBC were 97.40% ± 0. 62% and 0. 40%±0.15% respectively. 目的 建立流式细胞术快速鉴定ABO血型亚型的实验方法。方法 用荧光素(FITC)标记多人份人源抗-A、抗-B,与受检RBC上的相应抗原结合,利用流式细胞仪快速、特异地分辨每个细胞(生物颗粒)获得细胞的散点图,以RBC设定“门”,用Cell quest verl 2.2软件分析,对照管作“mark”,得测定管阳性细胞的百分率。结果 A型RBC其抗A-FITC百分率为97.40%±0.62%,抗B-FITC百分率为0.40%±0.15%;B型RBC其抗A-FITC百分率为0.42%±0.14%,抗B-FITC百分率为98.36%±0.55%。阴性细胞的x±s=0.43%±0.13%,阳性界值>0.85%(>x±3s)。11例ABO亚型FCM测定结果与血清学方法确认的结果完全一致。结论 流式细胞术鉴定ABO亚型的方法,较之常规的血清学方法,具有简便、快速、结果客观、重复性好之优点。
