After transforming the recombinant plasmids in BCG (Shanghai strain) by electroporation, the genomic DNA of all 21 transformants were prepared and hybridized with OmpL1. It showed that 6 of the 21 transfomants were recombinants in which the OmpL1 gene had been integrated into the genome of BCG.By immunoblotting with OmpL1 infected rabbit antiserum, which was preabsorbed to remove antibody against E. coli and SPAHRP, three recombinants, pLI1, pLI2 and pLI3, were detected to express OmpL1 protein. The ability of expression is in the order of pLI2>pLI1>>plI3. These studies provide the possibility of further research on the development of highly efficient recombinant vaccines against leptospirosis.

采用PCR技术直接从致病性钩端螺旋体(简称钩体)赖型017菌株基因组中扩增钩体外膜蛋白基因OmpL1,并克隆到大肠杆菌—卡介苗穿梭质粒载体pY6002中,重组质粒经电转化导入卡介苗。斑点杂交筛选重组卡介苗,再通过免疫印迹对其表达进行初步研究。在所得6个重组卡介苗中,有3个表达了OmpL1基因产物,其中一个表达较强。该研究为发展新一代高效广谱的钩体基因工程疫苗打下了基础。

目的 将表达IL 2重组BCG用于动物实验 ,观察其对肿瘤生长和机体免疫功能的影响。方法 采用基因工程技术和电转化技术 ,构建重组卡介苗即rBCG IL 2和rBCG ,并用瘤株重组BCG接种荷瘤小鼠 ,几周后观察肿瘤生长情况和检测巨噬细胞的杀伤活性。结果 rBCG IL 2能表达分泌IL 2 ,其表达量为 9 87ng ml± 4 56ng ml,rBCG IL 2与rBCG抑制肿瘤生长的作用明显优于对照组 ,并能提高小鼠巨噬细胞杀瘤率。结论 rBCG IL 2能有效抑制肿瘤生长 ,较早刺激机体的免疫功能 ,提高巨噬细胞的杀瘤活性。

Methods: Gene engineering technology, electricity transfer- ence and inoculation tumor cell are adopted; MTT assay is used to determine tumoricidal effect of macrophages. Results: Tumo- ricidal effect of macrophages of rBCG-IL-2 groups was higher than those of the control coups and BCG groups, and the tumorici- dal effect of macrophages could reach a high state in a week. Conclusion: rBCG-IL-2 can raise tumoricidal effect of macrophages, improve immunity of host and is a prospect biological preparation.

目的:研究能分泌人白细胞介素2的重组卡介苗(rBCC-IL-2)对小鼠腹腔巨噬细胞体外杀伤活性功能的改变,探讨rBCG-IL-2对宿主免疫功能的影响。方法:采用基因工程技术和电转化技术构建卡介苗,采用MTT法测定小鼠腹腔巨噬细胞体外的杀伤活性。结果:rBCG-IL-2组小鼠腹腔巨噬细胞杀伤活性明显大于rBCG组和对照组,且巨噬细胞的杀伤活性在一周内达到最大。结论:rBCG-IL-2能提高巨噬细胞的杀伤活性,是一种很有前景的生物制剂。
