Amylose content in rice grain was determined with Simplified Iodine-Blue Colorimetry(SIC) and State Standard Colorimetry(GB/T 15683-1995) at a temperature gradient of 15,20,25℃,and the paired data were subjected to t test.The determination results of the two methods were not significantly different at different gradients.At 15℃,amylose content was slightly higher as determined with SIC than that determined with GB when the content was low,and an opposite picture was true when the content was high.At 20,25℃,the measured value with SIC was generally higher than that with GB.Therefore,the difference between the two methods was slightest at 15℃.In addition,the measured value with SIC seemed to be fairly stable at different temperatures,with little variation in the slope of the standard curve.It is thus recommended that SIC be adopted for amylose determination at 15℃ for rice grain.

在15,20,25℃3个温度梯度下,利用简易碘蓝比色法与GB/T 15683-1995分别测定稻米直链淀粉含量,测得的结果作成对数据的t测验发现,在15,20,25℃下,差异均不显著。在15℃下,当直链淀粉含量较低时,简易法的测定结果略高于国标法,而直链淀粉含量高时则相反.在20,25℃下,简易法的测定结果普遍高于国标法,因此,2种测定方法在15℃下差异最小。通过研究还发现,简易法在不同温度下的测定值较稳定,标准曲线的斜率变化不大,因此简易法在温度15℃左右可以用于测定稻米直链淀粉含量,高于20℃则用简易法测得的结果由于普遍偏高,因此可用于粗略的估计,不宜作为精确的测定方法。
