Olaquindox is an antimicrobial veterinary additive with a long history of use as a growth promoter in stock raising. Although this drug has been widely used for its low price and convenient usage, olaquindox is actually still under evaluation because of the uncertainty regarding the mechanism involved in the slight increase in the incidence of adrenal cortical adenomas in male mice and the genotoxic potential of olaquindox. In this study, the zebrafish (Danio rerio) acute toxicity test, the zebrafish embryo test and the single cell gel electrophoresis test (SCGE, comet assay) of crucian (Cyprinus carpio) kidney cells in vivo were conducted to evaluate the toxicological effects of olaquindox on typical aquatic species. The result of the acute toxicity test showed that the 96 hrLC_(50) of olaquindox was 1 996.87 mg/L and the toxicity of olaquindox was very low according to the national aquatic spieces test manual. The result of the embryo test showed that the 96 hrEC_(50) of olaquindox was 221.20 mg/L and some of the tested embryos appeared to be badly affected by olaquindox, especially at high doses of exposure. The results of the SCGE showed that olaquindox could induce DNA damage in crurian kidney cells and might have a potential genotoxicity. The damage caused by olaquindox was obviously great and the diference was extremely significant, compared with those of control (P<0.01). The results of this study indicated that the use of olaquindox as a veterinary drug should be carefully restricted to ensure the safety of food and the well_being of mankind.

兽药添加剂喹乙醇是一种在畜禽及水产养殖中应用广泛的促生长剂 ,但其安全性仍存在较大争议 ,使用不当或过量使用会对环境构成潜在危害 .采用斑马鱼和鲤鱼作为生物材料 ,分别研究了兽药添加剂喹乙醇对斑马鱼的急性毒性、对斑马鱼胚胎发育的影响及对鲤鱼肾细胞DNA的损伤 .急性毒性试验表明 ,96hrLC50 为 1 996.87mg/L ;胚胎发育试验实验结果表明 ,96hrEC50 为2 2 1 .2 0mg/L ;单细胞凝胶电泳试验结果表明 ,喹乙醇能引起较大的DNA损伤 ,与空白对照相比有极显著性差异 (P <0 .0 1) ,并在设定的剂量范围内呈现一定的剂量效应关系

采用斑马鱼、鲤鱼作为生物材料,通过斑马鱼急性毒性试验、胚胎发育试验和鲤鱼肾细胞单细胞凝胶电泳试验,分别从生物个体水平、细胞水平和分子水平,对兽药及饲料添加剂喹乙醇、阿散酸和土霉素进行了生态毒理学效应研究。急性毒性试验结果表明,3种兽药及饲料添加剂在有生态学意义的浓度范围内对斑马鱼的急性毒性均不大;胚胎发育试验结果表明,喹乙醇和阿散酸对斑马鱼胚胎发育72 h的EC50值分别为221.20和239.54 mg.L-1,具有明显的致畸效应;单细胞凝胶电泳试验结果表明,喹乙醇和土霉素均具有一定的遗传毒性,能引起鲤鱼肾细胞DNA的明显损伤,并呈现出一定的剂量-效应关系。

The ecotoxicological effects of the veterinary additive olaquindox was studied,using zebrafish(Danio rerio),carp(Cyprinus carpio),and earthworm(Eisenia foetida)as experimental materials.The acute toxicity tests of fish and earthworm,the zebrafish embryo test,the single cell gel electrophoresis tests(SCGE,the comet assay)in vivo of carp kidney cells and earthworm coelomocytes were conducted,and the effect of olaquindox on cellulase and superoxide dismutase(SOD)of the earthworm Eisenia fetida,was investigated.The acute toxicity of olaquindox was not significant at environmentally relevant concentrations.The 72 h EC50 of olaquindox was 221.20 mg·L-1,and showed a significant teratogenic effect on the embryo.Olaquindox significantly increased the activity of cellulase and SOD at the concentration of 500 mg·L-1(P<0.05),and the activity of cellulase and SOD increased with the increasing concentration.The results of the SCGE showed that olaquindox could induce serious DNA damage obviously in both of carp kidney cells and earthworm coelomocytes in vivo,and the difference was very significant compared with the control(P<0.01),suggesting that olaquindox had potential genotoxicity to aquatic and edaphic animals.

采用水生生物斑马鱼(Daniorerio)、鲤鱼(Cyprinuscarpio)和土壤生物赤子爱胜蚓(Eiseniafoetida)作为生物材料,通过斑马鱼急性毒性试验、斑马鱼胚胎发育试验、鲤鱼肾细胞单细胞凝胶电泳试验、蚯蚓急性毒性试验、蚯蚓生化毒性试验以及蚯蚓体腔细胞单细胞凝胶电泳试验,对兽药添加剂喹乙醇分别从生物个体、细胞和分子水平进行了生态毒理学效应研究。急性毒性试验结果表明,喹乙醇实验浓度范围内对斑马鱼和蚯蚓的急性毒性均不大。胚胎发育试验结果表明,喹乙醇对斑马鱼胚胎的72hEC50为221.20mg·L-1,具有明显致畸效应。蚯蚓生化毒性试验表明,喹乙醇能促进蚯蚓体内SOD酶活性,并呈现一定剂量-效应关系,但对纤维素酶影响不明显,单细胞凝胶电泳试验结果表明,喹乙醇具有一定遗传毒性,能引起鲤鱼肾脏细胞和蚯蚓体腔细胞DNA损伤,与空白对照相比有显著差异,并呈现出一定剂量-效应关系。
