AIM: To compare the renoprotective effects of angiotensinⅡtypeⅠreceptor antagonist (AT 1RA) with that of angiotensin converting enzyme inhibitor (ACEI) and to investigate their influences on intrarenal renin-angiotensin system. METHODS: Experimental nephrotic syndrome model was induced in SD rats with repeated peritoneal injections of puromycin. Twenty-eight rats were randomly divided into four groups: normal control, nephrotic control, ACEI-treated and AT 1RA-treated group. Serum, urine and renal tissue were collected for study 12 weeks later. RESULTS: The urine protein was less and renal function was better in both treated groups. The glomerular and interstitial injury indexes of both ACEI and AT 1RA treated rats were lower than that of nephrotic control rats and had no significant difference between the two treated groups. The renal local ACE activity and angiotensinⅡ of nephrotic control group were significantly higher than that of normal control group and the two treated group(P<0.01). CONCLUSION: AT 1RA and ACEI have comparable renal protective effects on nephrotic syndrome and these effects may be related to the inhibition of renal local ANGⅡ.

目的 :比较血管紧张素受体拮抗剂 (AT1RA)和血管紧张素转换酶抑制剂 (ACEI)的肾脏保护作用 ,观察这两类药物对肾脏局部肾素 -血管紧张素系统的影响。方法 :肾病综合征模型由反复给SD大鼠腹腔注射嘌呤霉素诱导而成。2 8只大鼠随机分为 4组 :正常对照组、肾病对照组、AT1RA治疗组和ACEI治疗组。 12周后收集血、尿和肾组织标本进行检测。结果 :两治疗组尿蛋白明显少于肾病对照组 ,且到实验期末 ,肾功能仍在正常范围 ,肾小球和肾间质损伤指数也低于肾病对照组 ,但两治疗组间无明显差异 (P >0 0 5 )。肾病对照组的肾组织ACE活性、血管紧张素Ⅱ浓度显著高于正常对照组(P <0 0 1)。在ACEI和AT1RA治疗组 ,肾组织ACE活性和ANGⅡ浓度显著低于肾病对照组 (P <0 0 1)。结论 :AT1RA和A CEI在肾病综合征进行性肾损伤中 ,具有相似的肾脏保护作用 ,这种作用可能与抑制肾脏局部的ANGⅡ有关

The renal paraffin sections were stained with HE, PASM and ?-SMA immunohistochemical methods. Results In the control group,marked renal interstitial injury was noted,expression of ?-SMA was found in renal tuberlar epithelial cells and in their mesenchyma, whereas the above changes were much lighter in the group treated with Hong-Jing-Tian capsulae ( P < 0. 01 ). Conclusions ?-SMA was a good marker of MFB, which played an important role in lubulointerstitial fibrosis caused by unilateral urethral obstruction( UUO). Hong-Jing-Tian capsulae had a effect against renal interstitial fibrosis,being renal protective.

目的 探讨复方红景天制剂对肾间质损伤的保护作用。方法 制作由大鼠单侧输尿管梗阻(UUO)所致的肾间质纤维化动物模型。以α-平滑肌肌动蛋白(α-SMA)作为肌纤维母细胞(MFB)的特异性标志物,观察肾小管上皮细胞及间质内MFB的表达情况。26只SD大鼠随机分成假手术组、对照组和口服复方红景天制剂组,10天后全部处死,取肾组织采用HE,PAS及免疫组织化学α-SMA染色,行病理学及组织化学观察。结果 在实验结束时对照组肾间质病变显著,肾小管上皮细胞及间质α-SMA表达明显增加,而口服复方红景天制剂组则明显轻于对照组(P<0.01)。结论 α-SMA是MFB较好的标志物,MFB在UUO导致的肾间质纤维化中扮演了重要角色。复方红景天制剂具有抗肾间质纤维化的作用,对肾间质损伤有较好的保护作用。
