Two functional parameters evaluated by NBT dye reduction and YC rosette formation and a morphological change assessed by Wright-Giemsa stain of HL-60 cells were used to determine the extent of HL-60 cell differentiation induced by retinoids and DMSO. Two functional parameters evaluated by NBT dye reduction and YC rosette formation and a morphological change assessed by Wright-Giemsa stain of HL-60 cells were used to determine the extent of HL-60 cell differentiation induced by retinoids and DMSO. Studies revealed that under the action of retinoic acid(RA) and its two new synthetic analogs, 4-(ethoxycarbophenyl) retiamide(RI) and 4-(hydroxycarbophenyl) retiamide (RII), HL-60 cells could be induced to differentiate to more mature cells with functional characteristics of granuloeytes. When HL-60 cells were incubated with 10~(-6)M of RⅠ and RⅡ for 6 days, 42.3% and 43.6% of the eells were capable of reducing NBT; 20.7% and 14.9% could form YC rosettes with yeast ceils covered by human C_3 complement. DMSO was also shown to be active in the induction of both functional differentiation phenotypes. However, the expression of C_3 complement receptor was much earlier than the emergence of NBT reduction capacity under its induction. It is resonable to speculate that the induction machanism of retinoids and DMSO is somewhat different. Both retinoids and DMSO could induce HL-60 cells into morphologically more mature myeloid cells including myeloeytes, metamyelocytes and banded neutrophils. However, only a few cells matured into fully segmented ones. The induction of differentiation of HL-60 cells by all of the drugs tested was found to be associated with a reduction of the number of viable ceils. Since this phenomenon appeared later than cell differentiation, it may be related with the change of growth characteristics of differentiated cells.

The HL-60 cell line grew in liquid cultures as separate cells. HL-60 cell line had a doubling time of 25~31 hr with a cell saturation density of about 20×10~5m1~(-1). The morphology of most of the HL-60 cells resembles promyelocyte. The HL-60 cells reacted strongly with cytochemical stains specific for myeloid cells,including naphthol AS-D chloroaceta te esterase, myeloperoxidase,and Sudan black B.These cells did not react or weakly reacted with α-naphthyl acetate esterase (α-NAE) stain. Uninduced HL-60 cells did not reduce nitroblue tetrazolium (NBT) dye. The HL-60 cells were capable of forming colonies in the soft agar semi-solid culture without addition of any stimulating factor. When HL-60 cells were incubated with TPA for 1~4 days, the percentage of cells adherent to plastic or glass substrate and α-NAE-positive increased. The shape of differentiated cells became macrophage-like. The extent of HL-60 cell differentiation induced by TPA paralleled with concentrations of the compound which were 0.162~162 nM. On the other hand, HL-60 cells exposed to 1000 nM RA for 2~6 days differentiated into more mature myeloid cells including myelocytes, metamyelocytes, band and segmented neutrophils. The percentage of the cells capable of reducing NBT dye increased, it could reach 90% at 6th day after treatment with RA. The induction of differentiation by RA was concentration-dependent too, its effective concentrations were 1~1000nM. The proliferation of HL-60 cells was inhibited while differentiation was being induced by both TPA and RA.

实验观察了HL-60细胞的一些生物学特点及TPA和RA的分化诱导作用。HL-60细胞在液体培养中呈单个细胞悬浮生长,倍增时间25~31h,饱和密度20×10~5/ml左右。HL-60细胞形态类似早幼粒细胞,具有早幼粒细胞的细胞化学特征,NBT还原反应阴性或弱阳性。HL-60细胞在软琼脂培养中能自发形成集落。TPA诱导HL-60分化为巨噬样细胞。TPA与RA的分化诱导作用均与药物浓度有依赖关系,有效浓度分别为0.162~162nM和1~1000nM。TPA与RA诱导HL-60细胞分化的同时,对细胞增殖有抑制作用。
