Wheat fusarium head blight(scab) causes severe worldwide problems.It is likely that the fungi(Fusarium graminearum) that cause scab use alkaline protease when they invade grains.Purification of the enzymes is important to detect the mechanism which the fungi use to hydrolyze grain protein.To do this,an optimization test concerning the fermentation conditions,such as carbon sources,nitrogen sources,initial pH,culture time,inoculum size,inoculating time,was conducted with the culture strain F.graminearum F15.The results showed that the optimal alkaline-protease-producing activity of 534.2 U/ml was reached 84 h after culturing in a rotary shaker(120 r/min) at 28 ℃ with the medium containing 3.000% glucose,2.000% peptone,2.500%bran,0.100%KH_(2)PO_(4),0.200%KCl,0.001%ZnSO_(4),0.001%CaCl_(2),and under culture conditions of initial pH 9.0,8% inoculum size and 60 h inoculating time.

小麦赤霉病是全球毁灭性作物病害之一。禾谷镰刀菌产生碱性蛋白酶与侵染麦粒有关,纯化碱性蛋白酶对研究真菌降解谷物蛋白有重要作用。本研究探讨了培养基及发酵条件对禾谷镰刀菌菌株F15产碱性蛋白酶的影响。试验结果表明,优化的禾谷镰刀菌产碱性蛋白酶的液体发酵条件为:培养基含葡萄糖3.000%、蛋白胨2.000%、麸皮2.500%、KH2PO40.100%、KC l 0.200%、ZnSO40.001%、CaC l20.001%;培养条件为:初始pH9、接种量8%、种龄60 h,在120 r/m in下28℃振荡培养84 h,碱性蛋白酶活力达534.2 U/m l。
