Quantitative reversed phase high performance liquid chromatographic method was developed for the separation and determination of rutin and quercetin from mulberry leave extract. The instruments used were P100 HPLC system and DL 80 chromatographic work station. The column used was YWG C 18 ,10μm,250mm×4.6mm i.d. and eluent was MeOH H 2O H 3PO 4 solution (60∶40∶0.4). A good linear relationship of calibration curves(for rutin and quercetin) was found. The coefficients of variation for rutin and quercetin...

The recoveries of the two components were 104.0% and 103.2%, respectively.

用P100型高效液相色谱系统、DL-800色谱工作站、YWG-C18色谱柱,以甲醇-水-磷酸体系(60∶40∶0.4,V/V/V)为流动相分离并同时测定了桑叶提取物中芸香甙与槲皮素的含量。芸香甙、槲皮素含量与峰面积呈良好的线性关系。芸香甙的变异系数为1.98%,回收率为104.0%;槲皮素的变异系数为9.8%,回收率为103.2%。

In this study, Folium Mori,Radix scrophulariae, Radix Paeoniae Alba,Pericarpium Citri Reticulatae,Semen Armeniacae Amarum and Indigo Naturalis in Liyan Zhike Tablet were identified by thin-layer chromatography. The paeoniflorin content of Radix Paeoniae Alba was analyzed quantitatively by HPLC. It was shown that the methods are sensitive with good reproducibility, the rate of recovery is 96.66% and the RSD is 2.88%(n=5).

Rutin and quercetin are the main effective components of mulberry leaves with the functions of controlling the increase of fat in serum and controlling the formation of arterio sclerosis In this article a high performance capillary electrophoretic(HPCE) method was used to separate and determine rutin and quercetin in mulberry leaves collected from different periods, different places and different kinds in Xinjiang Electrophoretic conditions were as follows: a capillary tube (75 μm i d ×57 cm(effective length,50 cm)), with 10 mmol/L dihydrogen sodium phosphate 20 mmol/L sodium borate containing 15% methanol as the running buffer (pH 8 62) and an applied voltage of 20 kV, at 25 ℃, detected at a wavelength of 245 nm Under the optimum conditions, rutin and quercetin were separated successfully from other components within 12 minutes The corrected peak areas of rutin and quercetin increased linearly with the increase of their concentrations in the range of 4 4 mg/L 28 8 mg/L and 1 8 mg/L 145 8 mg/L respectively The corresponding regression equations for rutin and quercetin were Y=71 60ρ+30 51 (r =0 999?1 ) and Y=315 17ρ-929 70 (r =0 994?9)respectively, and the recoveries were 95.64% and 99.36% The analytical results demonstrate the method is simple, quick and well reproducible, and can be used as a reliable tool for the quality control of mulberry leaves

采用高效毛细管电泳法分离测定了新疆不同地区、不同采集期、不同品种的桑叶中的黄酮类成分芦丁、槲皮素的含量。以含有体积分数为 15 %甲醇的 10mmol/L的磷酸二氢钠 2 0mmol/L的硼砂溶液 (pH 8 6 2 )为电泳缓冲液 ,采用压力进样方式 ,在 2 5℃ ,2 0kV恒压下进行电泳分离 ,并在 2 45nm波长处检测。结果表明 ,桑叶中的两种目标组分在 12min内完全分离 ,且有良好的线性关系 ;芦丁和槲皮素的加样回收率分别为 95 6 4%和99 36 % ,其RSD分别为 2 2 5 %和 1 79% (n =6 )。方法简单、准确、快速。
