The natriuretic and diuretic effectsof atrium extracts of rabbits with con-gestive heart failure were found to begreatly decreasing as compared with con-trols (P<0.01), and observation fromelectron microscope indicated that thenumber of atrial specific granules,whichcontain cardionatrin,in congestive heartfailure rabbits also decreased as comparedwith that in normal rabbits.Both resultsanggest that in congestive heart failurethere is a decrease of cardionatrin inatrial tissue,and this may be due toits greatly...

Neriperside is a mixed glycoside containing mainly peruvoside, neriifolin and cerberin. The cardiac hemodynamics of peruvoside and neriifolin in anesthetized dogs, the biological activity and the safety margin of neriperside and its constituents in cats and guinea pigs were studied and compared with those of ouabain, digoxin and desJanoside. Neriperside is a mixed glycoside containing mainly peruvoside, neriifolin and cerberin. The cardiac hemodynamics of peruvoside and neriifolin in anesthetized dogs, the biological activity and the safety margin of neriperside and its constituents in cats and guinea pigs were studied and compared with those of ouabain, digoxin and desJanoside. Their pharmacokinetics were studied by ra-dioimmunoassay. The results indicated that the cardiac performance was improved during intravenous injection of neriifolin and peruvoside or intravenous infusion of neriperside, peruvoside, neriifolin, cerberin, digoxin,deslanoside, ouabain and strophan-thin K. According to therapeutic dose in guinea pig, the sequence of potency of their cardiotonie effect was peruvoside neriifolin neriperside deslanoside digoxin. The cardiotonic potency of cerberin was only a quarter of that of neriifolin.After intravenous injection of peruvoside or neriifolin, the plasma concentration curve was shown to fit a two compartment open model with the following pharma-cokinerie parameters :neriifolin T1/2α=11 min, T1/2β=6.79 h, Vd = 6.78 L/kg and CL = 0.69 L/h/kg; peruvoside T1/2α = 8 min, T1/2=2.13 h, Vd = 3.94 L/kg and CL=1.28 L/h/kg. After oral administration of peruvoside, neriifolin and neriperside, absorption was shown to be rapid with peak blood levels at 52 min, 34 min and 15 min respectively. The bioavailability of peruvoside, neriifolin and neriperside was found to be 29.5%, 35.6% and 27.1% respectively.

By orthogonal design, Zhenwu decoction are studied on the dogs and rabbits with experimental cardiac failure. The results show that the compatibility of the prescription with the original ingredients is most effective. The decoction can strengthen the contraction of cardiac muscle, improve the blood oxygen supply of the ischemic cardiac muscle, and promote the circulation of blood. 用正交设计法研究真武汤对在体犬及家兔实验性心力衰竭的作用,结果表明,真武汤原方组合是最佳配伍,能显著提高心衰犬和家兔的心肌收缩力,改善缺血心肌的血氧供应,促进血液循环,而对心肌耗氧量和传导系统无明显影响。方中配伍赤芍优于白芍,生姜似有触媒样作用,能增强附子的强心效力,又可相对减低附子的毒副作用。
