Biopsy of transitional cell carcinoma of urinary bladder taken before and after photodynamic therapy (PDT) from 15 patients were examined by electron microscopy. It has been demonstrated that this tumor is highly sensitive to PDT using hematoporphyrin derivative (HPD, China-made) and laser irradiation. Within the tumor tissues, the capillaries are most sensitive to PDT action, displaying distinct injuries of endothelium immediately after 20 minutes irradiation. 24 h after PDT, almost all capillaries became necrotic....

The cancer cells are less sensitive to PDT, showing destruction later and less serious as compared with that of capillaries. 4 days after PDT, the cancer tissue became completely necrotized. 3 weeks and 80 days after PDT, the tumor bed was still covered with necrotic debris. It is concluded that in PDT treated tumors, the capillary endothelium will be primarily damaged while destruction of cancer cells be mainly caused by structural and functional changes in microcirculation.

电镜观察15例光动力学治疗前后膀胱移行细胞癌的活检材料,证实此治疗对膀胱癌有显著作用。肿瘤组织内的毛细血管对光动力学作用最为敏感,照光20min后即出现明显损伤,24h后,毛细血管几乎完全坏死崩解。癌细胞的破坏出现较晚,迟于毛细血管。治疗4d后肿瘤组织水肿、出血,几全部坏死。治疗21~80d后,瘤床仍水肿并有出血及坏死物质复盖。表明血管的结构损伤与功能障碍在肿瘤光动力学治疗机理中起主要作用。

With HPV_(16,18) and HPV_(11) DNA probes, 80 cervical lesions and 6 other tissues were done to study human papillomaviruses (HPV) DNA sequences by dot blot hybridization. It showed that the positive rates of HPV_(16,18,11) DNA sequences in cervical cancer, cervical dysplasia and chronic cervicitis were 44% (22/50) , 8% (4/50) and 4% (2/50); 15% (2/13), 15% (2/13), 15%(2/13) and 19%(3/16), 0 and 13 (2/16) respectively. 3 of these positive samples showed weak cross-hybridization between HPV_(16) and HPV_(18)....

应用HPV_(16,18,11)三个DNA探针,通过斑点杂交技术,对80例宫颈疾患和6例其它组织进行了检测和分析,结果显示HPV_(16、18)和HPV_(11)DNA序列在宫颈癌、宫颈不典型增生和慢性宫颈炎中的检出率分别为44%(22/50),8%(4/50),4%(2/50);15%(2/13),15%(2/13),15%(2/13)和19%(3/16),0%(0/16),13%(2/16);其中有3例标术HPV_(16,18)呈弱交叉杂交。而1例子宫肌瘤、1例阴道癌、1例宫颈息肉、2例膀胱癌和2例正常子宫颈组织中均未测出HPV DNA序列。结果提示:某些HPV类型(特别是HPV_(16))可能与宫颈癌的发生有关。

Cytokeratin and Carcinoembryonic Antigen (CEA) in bladder cancer cells were studied in 28 and 26 cases respectively using Avitin-Biotin-peroxidase Complex (ABC) method. The results showed that the low molecular weight cytokeratin and CEA expressed during the embryo development were also expressed in transitional cell carcinomas. Cytokeratin and Carcinoembryonic Antigen (CEA) in bladder cancer cells were studied in 28 and 26 cases respectively using Avitin-Biotin-peroxidase Complex (ABC) method. The results showed that the low molecular weight cytokeratin and CEA expressed during the embryo development were also expressed in transitional cell carcinomas. Both were increased with the increase of pathological grade and de- creased with degree of cancer cell differentiation. It is suggested that tha differences of both cytokeratin and CEA in the different pathological grading of bladder car- cinoma might be an index related to the differentiation of cancer cell. The results also showed that by using higher sensitive method and specific antibody, the antigen of cancer cell may be demonstrated well in formalin-fixed, fresh or older, paraffin- embedded tissues.

本文应用ABC免疫过氧化酶技术分别对28例和26例膀胱癌细胞角蛋白和癌胚抗原进行了检查。结果表明,移行细胞癌随着细胞分化程度的降低,于胚胎发育过程及正常上皮细胞成熟过程早期表达的部分基因,在膀胱癌细胞内重新表达或表达失常,从而影响细胞的分化;不同病理组织学分级的膀胱癌细胞内角蛋白含量的差异,可作为癌细胞分化程度的一项指标。本文还提出,应用高敏感性的方法及高特异性抗体仍可较好地检测出福尔马林固定、石蜡包埋的新鲜或陈旧的组织中癌细胞的抗原性。
