Objective To explore the synergistic effects of nerve growth factor (NGF) and ciliary neurotro-phic factor (CNTF) on the recovery of electrophysiologicai function in peripheral nerve regeneration (PNR) with the new cabin model for PNR. Methods Thirty-six Sprague-Dawley rats had a 10 mm gap in their sciatic nerves, bridged with a new double channel nerve conduit in fusiform shape. They were randomly divided into three groups (n = 12): group A (200 μl of chitin) , group B (100 μl of chitin and 5 μl of NGF or NGF and CNTF) and group C ( 100 μl of chitin and 5 μl of CNTF or NGF and CNTF). The morphological and pathological changes in any regenerated nerves were observed at 8 and 12 weeks after the operation. Compound muscle action potential (CMAP) , cortical somatosensory evoked potential (CSEP) and conductive velocity (CV) were observed as well. Results There was no significant difference of morphology or pathological function between the two sides in group A. The regenerative fibres were welldistributed and arrayed more regularly than those in the control, while the latency of CMAP and CSEP was shorter and the amplitude was higher (P<0.05). Conclusion NGF and CNTF might have synergistic effects on the recovery of electrophysiologicai function in peripheral nerve regeneration.

目的利用新型神经再生小室模型探讨联合应用神经生长因子(NGF)和睫状神经营养因子 (CNTF)促进周围神经电生理功能恢复的效果。方法用自行研制的梭形双通道桥接管桥接36只大鼠坐骨神经10 mm缺损,根据梭形管的两支管内注入的药物不同将动物随机分为A组(两支管内均注入几丁糖凝胶)、B组(一侧支管内注入几丁糖+NGF,另一侧支管内注入几丁糖+NGF+CNTF)和C组(一侧支管内注入几丁糖+CNTF,另一侧支管内注入几丁糖+NGF+CNTF)。于术后8,12周对再生神经的大体形态、组织病理学进行观察,并对术后12周的再生神经行皮质感觉诱发电位(CSEP)、复合肌肉动作电位(CMAP)及神经传导速度(CV)测定。结果 A组两支管内再生神经纤维组织形态学及电生理功能差异无统计学意义。B 组和C组注入NGF+CNTF侧再生纤维粗细均匀,排列整齐,明显优于对照侧,CSEP、CMAP的潜伏期短、振幅高、传导速度快,两者差异有统计学意义(P<0.05)。结论 NGF与CNTF对周围神经电生理功能的恢复具有协同作用。

Objective:To discuss the synergistic effects on regenerative nerve fibers affected by nerve growth factor(NGF)and ciliary neurotrophic factor(CNTF)with the new cabin of peripheral nerve regeneration.Methods:Thirty-six Sprague-Dawley rats with 10 mm gap of sciatic nerve were averaged divided randomly into three groups(A,B,C),which had been bridged with the new double channel nerve conduit of fusiform shape.In group A,100 μl chitin for medical use was injected into the two branches of the conduit;in group B,beside injection chitin,5 μl NGF and normal saline,5 μl NGF and CNTF were respectively injected into the two branches;in group C,beside injection chitin,5 μl CNTF and normal saline,5 μl NGF and CNTF were respectively injected into the two branches.At 8 and 16 weeks after operation,the morphology and ultrastructure of regenerative nerve were evaluated by HE staining and electron microscope.The function of axoplasmic transport was analyzed by nuclear yellow.Results:The regenerative nerve in the side where NGF and CNTF were injected were light yellow,and the resilient is also senior to the ones in the control side.The diameter of the regenerative nerve fibers in the synergistic side is coincident and line up in order under light microscope,the number of myelinated or unmyelinated fibers are more,the thickness of myelinated fibers is larger,the function of axoplasmic transport was also better than the nerve in thd control sides.Conclusion:There is significant synergistic effect of NGF and CNTF on promoting the regeneration of peripheral nerve.
