This study was done to establish a human lung adenocarcinoma cell line SPC-A1 with the characterization of docetaxel resistance, to investigate its biological mechanism of drug resistance and how to reverse the resistance.
Conclusions:The SPC-A1/Docetaxel cell line showed high resistance to docetaxel, and had the characteristic of multidrug resistance. The cell line could be useful for the study of drug resistant mechanism and its reversal of docetaxel.
2. The resistance cell line, ACC/VCR, was induced in the ACC cell line in vitro by progressive concentrations of vincristine(VCR), a drug of choice in the treatment of adenoid cystic carcinoma. 3. The growth curve ,drug sensitivity in the parental and resistant cell lines were investigated by cell counting, flow cytometry and MTT assay.
Methods The serial passage tests were done in 7 sensitive Helicobacter pylori strains (2 type strains and 5 clinical isolates) to induce the resistance to amoxycillin, tetracycline, furazolidone, metronidazole or clarithromycin in vitro.
When the mice were treated with CIP at the same time,the rate was 34%. When strain 1 was induced to resist more drugs,the rate went up to 54% remarkably.
Susceptible and resistant strains after the in vitro induction of resistance were examined to evaluate mutations of the rdxA and frxA genes by sequence analysis.
After the in vitro induction of resistance, analysis revealed that two and four susceptible strains developed resistance when cultured with 0.3?μg/ml and 0.6?μg/ml of metronidazole, respectively.
The results after the in vitro induction of resistance to metronidazole suggested the presence of additional metronidazole resistance mechanisms, other than mutations of the rdxA and/or frxA genes.
