Studying the effect of Mifepristone (RU486) on ultrastructure of human trophoblastic cells during the early pregnancy, we found that RU486 caused the trophoblastic cells to dis-play morphological changes such as caryolysis, rough endoplasmic reticulum dilatation, mitochondria shrinkage and syncytial epithelium dissociation etc. Although RU486 is progesterone antagonist in receptor level, no apoptotic cell death was found in the trophoblastic cells. According to referred data, only degeneration or necrosis takes place in the decidua cells under RU486 effect. These findings indicated that the morphological changes both in the trophoblastic cells and decidua cells are resulted from secondary effect of RU486. The mecha-nism of RU486 to terminate the early pregnancy may be connected with the decrease blood supply of the utrine spiral arteries.

Methods Balb/c mice were immunized;specificity and cytotoxicity of the mAbs were determined by cell immunization technigue,cell ELISA and complenment dependent cytotoxicity(CDC),respectively. Aim To prepare monoclonal antibodies(mAbs)to tro phocytes and analyse their biological properties. Methods Balb/c mice were immunized;specificity and cytotoxicity of the mAbs were determined by cell immunization technigue,cell ELISA and complenment dependent cytotoxicity(CDC),respectively. The specificity, cytotoxicity and antigen binding sites of the mAbs were also investigated. Results These mAbs from 8 hybridoma cells had high specificity to trophocytes,two of them had cytotoxicity. They bound mainly to Mr 80 000 and 40 000 molecules of epitopes on trophocytes;The reaction of the mAbs to trophocytic antigen was blocked by the sera from immune abortion patients. Conclusion The antigen binding sites of the mAbs may be specific membrane antigen site of trophocytes which induce immune abortion.

目的研制抗人滋养细胞单克隆抗体并分析其免疫生物学特性。方法采用滋养细胞免疫Balb/c小鼠 ;用细胞ELISA法鉴定mAb的特异性 ;用补体依赖的溶细胞作用测定mAb的细胞毒性。结果共获得8株分泌抗人滋养细胞mAb的杂交瘤细胞 ,其中2株mAb具有明显的细胞毒活性。抗滋养细胞mAb主要针对相对分子质量 (Mr)为80000和40000的两组滋养细胞表面抗原。免疫性流产患者的血清 (含抗滋养细胞抗体 )可阻断mAb与滋养细胞膜抗原的反应。结论利用细胞免疫法所获该组mAb所针对的抗原表位 ,可能是诱发免疫性流产的滋养细胞膜特异性抗原。
