Authors modified the analytical method for the determination of the content of capsaicin in capsaicum.The extra of blank tube was used for the sake of elimination of color interferance;the concentration of solution of sodium carbo- nate was changed in order to moderate the operating condition and prevent precipitate from being appeared under low temperature.The modified method has many advantages:simplicity and convenience,high speed operation and excel- lence in reappearance.

A one-step method (Mayahara et al. .1980), using p-nitrophenylphosphate (p-NPP) as the substrate and lead citrate as the capture agent, ,was adopted to demonstrate the ouabain-sensitive, potassium-dependent p-nitrophenylphosphatase (K+-NPPase) catalyzing the second step (dephosphorylation) of the Na+-K+-ATP-ase system, and thus the Na+-K+-ATPase activity was cytoohemioally located. The present work showed that the reaction products were located on the saroolemma, T-tubules, sarcoplasmio reticulum and outer membrane of mitochondria in ultrathin sections of normal rat cardiooyles. A series of control experiments proved that this method was more specific, and nonenzymatio hydrolysis of ATP'by lead was not involved. The findings showed that isoproterenol (ISO) inhibited the activity of Na+-K+-ATPase, and verapamil protected the Na+-K+-ATPase activity from the Inhibitory effect of ISO.

We have prepared from different animals,namely SD rats,domestic suckings pig and acephalous newborns the hepatocyte extract that can stimulate hepatocyte DNA synthesis,by modified Labrecque method.The high performance liquid chromatography showed that the main component of the HSS we prepared was quite similar to that reported by Labrecque and no significant difference was found among the HSS from the three species of animals,Ultra-violet scanning revealed two main peaks of UV-absorbance in the samples supposedly...

We have prepared from different animals,namely SD rats,domestic suckings pig and acephalous newborns the hepatocyte extract that can stimulate hepatocyte DNA synthesis,by modified Labrecque method.The high performance liquid chromatography showed that the main component of the HSS we prepared was quite similar to that reported by Labrecque and no significant difference was found among the HSS from the three species of animals,Ultra-violet scanning revealed two main peaks of UV-absorbance in the samples supposedly due to the presence of much nucleic acid aside from the poly-peptide component.Its Mn and Mw were 11685D and 11340D respectively as estimated by average molecular weight determinations.The biological activity assays of the HSS by our modified method or the standard method confirmed the existance of the active substance in either extract and disproved the presence of insulin and glycogen.All these experimental results suggested that the modified mathod can replace the Labrecque method and the preparation of HSS from suckling pig liver instead of human fetal liver is applicable.The modified method is superior in its easier preparation and higher output with lower cost,therefore easily adopted in all laboratories.

作者改进了LaBrecque提取HSS的繁琐工艺(简称改良法),从SD大鼠、乳猪、无脑儿的新鲜肝脏制备出能促进肝细胞DNA合成的物质(Hepatic Stimulator Substance,HSS)。经高效液相色谱(HPLC)分析表明:改良法制备的HSS的主要组分与LaBrecque的HSS色谱行为基本相似,而且SD大鼠、乳猪、无脑儿之间无明显差异。紫外扫描发现HSS样品中有两个主要吸收峰,推测除多肽成份外还有较大比例的核酸存在。平均分子量测定:数均分子量为11685D,重均分子量为11340D。两种方法制备的HSS均能刺激肝细胞DNA合成,并排除有胰岛素,胰高糖素存在的可能性。几项试验结果表明,乳猪肝脏可以替代人胎肝提取HSS;改良法可以替代LaBrecque报告的方法,而且改良法操作简便、产量高、成本低、易于推广。
