Methods Rat kidney transplantation was performed according to the procedure of kamada, with some modification. Experimental rats were divided into 5 groups. The survival time of recipient rats and function of grafts after renal transplantation were observed. The sections of renal graft were stained for monoclonal antibody ICAM1 and LFA1, and then quantification of ICAM1 and LFA1 expression was accomplished by computer image analysis. Results ICAM1 and LFA1 increased significantly in the renal allograft rejection group as compared with the nonrejection group (P<0.05). Conclusion Both the biopsy of renal graft and the monitoring of ICAM1 and LFA1 are useful tools in diagnosing and treating acute rejection.

目的探讨移植肾组织内细胞间粘附分子-1(ICAM-1)/淋巴细胞功能相关抗原-1(LFA-1)的异常表达与移植排斥的关系。方法采用改进的大鼠原位肾移植模型,分五个实验组,在不同的时间段、观测受体鼠存活、肾功能;以及移植肾组织用单克隆抗体免疫组织化学染色后,图象分析法定量测定移植肾组织ICAM-1/LFA-1分子表达的水平。结果移植肾急性排斥组ICAM-1/LFA-1分子水平显著高于同品系移植对照组和药物治疗组。结论移植肾组织活检同时检测组织内ICAM-1/LFA-1分子的表达,在肾移植急性排斥的诊断和治疗方面具有极其重要的临床意义。

目的 找到预防大鼠原位肾移植模型建立过程中发生并发症的方法。方法 采用动脉端 侧吻合、静脉袖套吻合、输尿管端 侧吻合的方法进行 6 9次大鼠原位肾移植实验 ,并分析死亡的原因。结果 共有 6 1只大鼠存活 ,8只大鼠死于并发症。结论 大鼠肾移植手术的术前准备、术中操作及术后护理对模型的建立有重要影响 ,采取具体的保护措施可以减少大鼠的死亡率

Objective To study the relationship between erythrocyte immune function and SOD, MDA in renal orthotopic transplantation of rats with acute rejection.Methods The erythrocyte C3b receptor rate(C3bRR),immune complex rate(ICR),immune adherence enhance factor(RFER) and immune adherence inhibitor factor(RFIR) were determined in 20 renal transplanted rats with acute rejection and in 20 healthy rats.Sero superoxide dismutase(SOD) and the content of lipioxidatation malondiadehyde(MDA) were also determined.Results C3bRR and RFER were significantly lower( P <0.01),while ICR and RFIR were higher( P <0.01) in the transplantation group as compared with the healthy group.There were lower activites of SOD( P <0.01),and an obvious increase of MDA content( P <0.01), and a positive correlation between activites of SOD,MDA content and erythrocyte immune function.Conclusion Renal transplanted rats with acute rejection show lower erythrocyte immune function,lower activites of SOD and higher MDA content.There is a close relationship between the RBC immune function and activites of SOD, MDA content.It may be important to detect acute rejection in renal transplantation.

目的 对肾移植急排期间红细胞免疫功能和超氧化物歧化酶 (SOD)、丙二醛 (MDA)关系进行探讨。方法 建立大鼠原位肾移植模型 ,测定 2 0只术后急排期大鼠的红细胞C3b受体花环率 (C3bRR) ,免疫复合物花环率(ICR) ,免黏附促进因子 (RFER) ,免疫黏附抑制因子 (RFIR)及血清SOD酶活性、MDA浓度。并取 2 0只正常大鼠作对照组同步测定。结果 与对照组相比 ,急排组C3bRR、RFER显著降低 ,ICR、RFIR显著升高 ,且SOD酶活性降低、MDA浓度升高 ,并与红细胞免疫功能有相关性 (P值均 <0 .0 1 )。结论 肾移植急排期间红细胞免疫功能下降 ,SOD酶活性降低 ,MDA浓度升高且呈密切相关
