An immobilization procedure of lactoperoxidase by radiaion copolymerization and crosslinking was developed. We found that the native lactoperoxidase was very unstable under γ-ray rradiation. But the system of acrylamide and acrylic acid monomers and the enzyme was stable against 9×10~5R of irradiation dose, and could be converted to an enzyme gel in the frozen state at -20℃. Lactoperoxidase activity was over 50%. The optimum pH and temperature of the immobilized peroxidase was 5.4 and 40℃ respectively.The immobilized...

本文建立了利用辐射共聚交联制备固定化乳过氧化物酶的方法。在γ射线辐照下,液态乳过氧化物酶较易失活。但是酶与丙烯酰胺、丙烯酸混合单体的体系,在-20℃冷冻状态下,对9×10~5R的照射量却是稳定的,并且能转变成一种酶凝胶。乳过氧化物酶活力仍保持在50%左右。制得的固定化乳过氧化物酶的最适pH为5.4,最适温度为40℃。此固定化酶可用于催化放射性~(125)I标记胸腺素。
