The protein chip is based on both the high-density micro pore plates technique and DNA micro arraytechnique. With the protein chip, we can carry out the analysis of high throughput gene expression on protein level,which is becoming the most effective method in the research field of proteome. Through manual operation or press andspray, the protein probes were spotted on the chemical membrane, gel, micro pore plate or slide and form into arrays.The target protein can be bound via the hybridization with sample. Then using the atomic-force microscope, phosphorimaging system, optical density instrument or Laser Con-focal Scanner to examine the target proteins, we can obtaininformation about the mode and level of target protein expression. The protein chip has been used to study the constitut-ing and changing of protein expression profile, the interaction between protein and biomolecule(protein, nucleic acidand ligand), the screening of antigen and antibody, and the interaction between enzyme and its substrate. The proteinchip has potential applied value in the research field including clinical diagnose, curative analysis, drug screening.

在高密度微孔板技术和DNA微阵列技术的基础上发展起来的蛋白质芯片技术,能够在蛋白质水平上进行基因高通量表达分析,从而成为蛋白质组学研究的有效方法。蛋白质芯片依靠手工、压印或喷墨的方法将探针蛋白点样在化学膜、凝胶、微孔板或玻片上形成阵列,经过与样品的杂交捕获靶蛋白,再用原子力显微镜、磷光成像仪、光密度仪或激光共聚焦扫描仪进行检测,获得靶蛋白表达的种类、数量及关联等信息。蛋白质芯片已经用于研究蛋白质表达谱构成及变化、蛋白质与生物分子(蛋白质、核酸、配体等)的相互作用、抗原体筛选、酶与底物相互作用。蛋白质芯片在医学临床诊断、疗效分析和药物筛选方面具有潜在的重要应用价值。
