To determine whether there are different membranous antigem expressions among primary human hepatocellular carcinoma cells and hepatocytes, four intact human hepatoma cell lines namely BEL-7402, QGY-7703, MAH, SMMC-7721 as well as Hela cells were used to immunize New Zealand white rabbits. Antisera obtained were adsorbed on powdered human liver. Frozen sections from hepatocellular carcinoma (HCO) (33 cases), and non-cancerous liver tissue (10 cases) were stained by indirect immunofluorescence and PAP techniques....

To determine whether there are different membranous antigem expressions among primary human hepatocellular carcinoma cells and hepatocytes, four intact human hepatoma cell lines namely BEL-7402, QGY-7703, MAH, SMMC-7721 as well as Hela cells were used to immunize New Zealand white rabbits. Antisera obtained were adsorbed on powdered human liver. Frozen sections from hepatocellular carcinoma (HCO) (33 cases), and non-cancerous liver tissue (10 cases) were stained by indirect immunofluorescence and PAP techniques. The incidence of positive membranous staining of cancer cells among 33 cases of HCC using various antisera wero as follows: anti-BEL-7402, 78.8%; anti-QGY-7703, 69.7%; anti-MAH, 48.5,%; anti-SMMC-7721 and anti-Hela, 15.2%; but the cytoplasm and nuclei of HCC wore negative. The above membranous staining, of antisera treated jiver tissue and hepatocytes surrounding HCC or non-HCC hepatocytes were negative. In some cases of HCC and non-HCC liver tissues, weak staining of the sinus wall could be seen. Using the microspectrometer, 8 kinds of antisera treated sections of HCC and liver tissue using PAP staining technique were scanned for comparing the intensity of staining quantitatively in the same sections. The intensity was 2-17 times higher in HCG than in the liver tissues surrounding HCC. These findings suggest that there are different membranous antigen expressions between HCC and the surrounding liver cells.

The pathological features and the CPE of the tissue culture of the cervical cancer closely correlated with the frequency of HPV 16 DNA homologous sequences (P<0.001). It is demonstrated that HPV infection is closely related to cervical cancer.

本文报道应用Southern blot核酸分子杂交技术,以标记的国内HPV16型基因组分子为探针,共检测22例宫颈癌、4例宫颈不典型增生、6例宫颈慢性炎症,5例宫颈内膜息肉,6例正常宫颈上皮组织中的HPVl6型基因组同源序列。杂交结果表明,宫颈癌组织中HPV16型基因组同源序列阳性率为36.4%(相关序列阳性率为59.1%),非癌组织和正常宫颈上皮组织均为阴性,P<0.05具有显著性差异。同时还观察到宫颈癌病理组织学特征和组织细胞体外培养CPE与HPV16型基因组同源序列检出率呈正相关,P<0.001。更进一步证明宫颈癌发病与HPV感染密切相关。并提出HPV感染的初筛检测方法。
