The assay combined the high specificity of double-stranded displacing probes with real-time quantitative PCR.Using differently labeled probes, the relative amounts of each allele in a sample could be quantified in one reaction. This method was evaluated using CDs41-42(-TCTT) mutation in human β-globin gene causative for β-thalassemia as an example. The wild-type and mutant probes were labeled with FAM and ROX,respectively,and were recruited in one reaction. Using the constructed DNA pools, real time PCR was performed to establish the relationship between the relative amount of each allele and the threshold cycle value.With this relationship, allele frequency of the pooled DNA sample could be obtained by its threshold cycle value and the DNA concentration of the sample. The results showed that there is a linear relationship between the amount of the allele and the threshold cycle value through the whole allele frequency range studied,and the lowest allele frequency detected was 1%.The linear correlation coefficient was 0.997 7 for wile-type allele and 0.993 8 for mutant allele,respectively. Within the allele frequency range of 1%~90%,the relative error calculated was less than 4%,which was acceptable for allele frequency determination in pooled DNA samples.Considering its simplicity,reliability,and low cost,this approach was a powerful strategy for large population-based epidemiology study,detecting meaningful polymorphic differences in candidate gene association studies, and genome-wide linkage disequilibrium scans.

结合荧光双链置换探针的特异性和实时PCR的准确定量能力,建立了一种新颖、准确、价廉且高通量的测定DNA池等位基因频率的方法.该方法采用不同荧光标记的双链置换探针1次PCR反应即可测定出等位基因频率.实验以β 地中海贫血CDs41 42( TCTT)突变为对象,分别用荧光染料FAM和ROX标记野生型和突变型探针,由实时PCR检测建立的等位基因浓度与循环阈值的响应曲线计算DNA池的等位基因频率.结果表明,建立的系列等位基因浓度与循环阈值呈线性关系,线性相关系数分别为0.9977(野生型等位基因)和0.9938(突变型等位基因),可检测的最低等位基因频率达1%,等位基因频率在1%~90%范围内测定误差小于4%.该方法可广泛用于流行病学调查、遗传连锁分析以及全基因组连锁不平衡扫描等领域.

With CIRAS-2 portable photosynthesis system, diurnal variations and response to light of gas exchange parameters of clove (Syringa oblata Lindl.) leaf in four years old were studied under different soil moisture content in the loess hilly region in semi-arid area. The results indicated the Photosynthetic rate(P n), Transpiration rate(T r), Water Use Efficiency of Leaf(WUE), Stomatal conductance(G s) and Intercelluar CO 2 concentration (C i) had clear threshold value responses to soil moisture and Photosynthetic Active Radiation(PAR). The fitting range of Mass Water Content(MWC) is 15 0%~19 5%, Relative Water Content(RWC) is between 58 8% and 76 6%, the fitting PAR range is 600~1?000?μmol photons·m -2·s -1. In that range of MWC and PAR, Syringao blata lindl can get higher P n and WUE, and will not waste water because of high T r, and the P n and WUE will weaken out of those range. The lowest MWC is about 11 6%(RWC is 45 7%), and the corresponding highest PAR is about 800?μmol photons·m -2·s -1, the falling of MWC or the increasing of PAR will lead to serious aridity stress and high light stress. The results showed that in the hilly-gully region in semi-arid area, Syringa oblata Lindl. should be planted in the ubac rather than in the abret, or be planted under other trees to form mixed forest rather than pure forest, which may has higher productivity and effects.

在半干旱黄土丘陵沟壑区 ,应用英国PPS公司生产的CIRAS 2型光合作用系统 ,测定不同土壤水分下 4年生紫丁香 (SyringaoblataLindl.)叶片气体交换参数的日变化和光响应特性。结果表明 :丁香光合速率、蒸腾速率、叶片水分利用效率、气孔导度、胞间CO2 浓度等气体交换参数 ,对土壤水分和光合有效辐射的变化 ,具有明显的阈值响应。有利于丁香光合作用和水分有效利用的适宜土壤质量含水量范围在 15 %~ 19 5 %之间 ,土壤相对含水量为5 8 8%~ 76 6 % ;适宜的光合有效辐射强度范围在 6 0 0~ 10 0 0 μmol/ (m2 ·s)之间。在此土壤质量含水量和光合有效辐射强度范围内 ,丁香的光合作用和生长过程不会发生较大的水分胁迫和强光胁迫 ,也不会发生蒸腾速率过高造成的无效蒸腾耗水 ,因而能获得较高的光合速率和叶片水分利用率。维持丁香正常生理和生长过程所需的最低土壤质量含水量在 11 6 % (土壤相对含水量为 4 5 7% )左右 ,相应的最高光合有效辐射强度在 80 0 μmol/ (m2 ·s)左右 ,土壤质量含水量降低或光合有效辐射强度升高 ,会导致严...

The se conclusions do well to the clinic application of nerve electro-physiology; ne rve biology and neuron-threshold. Along with the experiment data enriched and co nsummated more and more, using analysis method for experiment, neuron-threshold will be up to par of clinic diagnosis and treatment.

神经阈值的检测实验使我们得到了不同神经纤维、不同刺激信号对于神经阈值响应的不同数据。如何从这些不同的数据中获得归一化的有规律的能够用以确定的数值。本文经过数据分析、统计学处理和频谱分析,确定了神经阈值归一化数值方法,初步找到与阈值参数相关的因素及其相互间的关系,证明了神经元非线性特性。这些结论为神经电生理神经生物学及其神经阈值的临床应用创造了良好条件。随着实验数据的不断丰富、完善,运用实验分析方法,神经阈值会愈来愈达到临床诊断和治疗的水平。
