Hyperlipidemia may cause renal damage and contribute to the developement of glomerular scleresis.We proved that Rheum Officinal (RO) could lower the plasma lipids and effectively retard the progression of chronic renal failure。In this study,we investigated the effect of low density lipoprotein (LDL) and Ernodin (a main component of RO) on the growth of rat mesangial cells (MC).It was found that LDL had dural effects on ~3H-TdR uptake of MC,stimulating at 25 mg/L, while at 125 mg/L or more dose-dependently inhibiting the ~3H-TdR incorporation of MC.Direct cytotoxic effect was observed when LDL at 500 mg/L by estimation of LDH release.We also found that Emodin inhibited the growth of MC,and strikingly antagonized the stimulatory effect of LDL at 25 mg/L.These results,together with our previously work,showed that LDL may participate in the pathogenesis of mesangium damage and Emodin is benificial to the prevention of glomerular sclerosis.

高脂血症可以导致肾脏损伤,促进肾小球硬化的发生.本文观察了低密度脂蛋白(LDL)对系膜细胞(MC)生长的影响,发现 LDL 对 MC 具有双向性作用。低浓度(25mg/L)促进系膜细胞~3H-TdR 掺入,浓度>1250mg/L 时则抑制 MC 生长,其抑制作用与浓度成正比.当 LDL 浓度达500mg/L 时对系膜细胞有直接损伤作用.LDL 对MC 的这两种作用均可能导致系膜的病变,大黄素可以抑制细胞~3H-TdR 掺入,并且抑制 LDL 对 MC 的刺激作用。可见大黄除了具有改善高脂血症的作用外,还能直接影响 LDL 对系膜细胞的损伤,从而有防治肾小球硬化作用.
