本实验以Percoll密度梯度离心法分离出大鼠肾皮质的近曲肾小管细胞(proximalconvolutedtubules,PCT)和远曲肾小管细胞(distalconvolutedtubules,DCT),应用荧光指示剂Fluo3-AM成功测定了人甲状旁腺激素[hPTH(1-84)]、双丁酰环磷酸腺苷酸(dbCAMP,系一种可进入细胞内的CAMP激动剂)及前列腺素E2(PGE2)对DCT细胞内钙浓度([Ca2+]i的影响,发现PTH、dbcAMP和PGE2均可引起[Ca2+]i的增加。提示:(1)cAMPP在DCT细胞中可能与PTH所致的[Ca2+]i的升高有关;(2)PGE2可能在PTH对[Ca2+]的调节作用中充当某种信使物质。
