This paper reports the effects of potassium chromate and dichromate onseven different genetic endpoints. Both these chemicals gave positive reacti-on in SCE, chromsome aberration, micronucleus and cell transformation inrat embrpo cell cultures, mitotic gene conversion assays in S. cerevisiae andinductive test of prophage(λ) in E. Coli. The positive responses were notobserved in mitotic chromsome diminution and reverse mutation assaysin S. cerevisiae. Based on these results a carcinogenicity prediction value(θ_8~+)...

Dose related functions indicate substantial quantitative agreementin results from the same tests performed using both chromium~(+6) and3-MCA(DRF = 2.08). The probability (P value) shows a significant matchfor chromium~(+6) and aflatoxin B that suggests concordant endpoints ingenotoxicity.

本文报道了铬酸钾与重铬酸钾对体外试验多个遗传终点的作用。其中在SCE、染色体畸变、微核分析、细胞转化、酵母菌有丝分裂基因转换和E.coli噬菌体λ诱导试验中,均获阳性结果,但未能诱导有丝分裂染色体丢失和回复突变。以此结果进行CPBS系统分析,其致癌性预测值(Q_8~+)为0.93,提示是潜在的致癌物;同时计算了比较概率P值和剂量相关函数DRF值,结果表明其作用终点与黄曲霉毒素B_1,3-MCA相似,作用强度与3-MCA相似。六价铬与砷,镉及三价铬等的作用有较大差别。

The experiments showed that (1) the expression process of Px genes in maize germinating embryo was identical to the germinating processes of maize seed embryos, whose expression sequencies and expression frequencies were all regulated genetically; (2) Px genes can be grouped into stable-expression type and variable-expression type, of which the expression patterns of the variable-expression type of Px genes can be converted definitely into specific manner " single-gene converter" or " double-gene converter"...

The experiments showed that (1) the expression process of Px genes in maize germinating embryo was identical to the germinating processes of maize seed embryos, whose expression sequencies and expression frequencies were all regulated genetically; (2) Px genes can be grouped into stable-expression type and variable-expression type, of which the expression patterns of the variable-expression type of Px genes can be converted definitely into specific manner " single-gene converter" or " double-gene converter" on the definite genetic background; (3) one of the causes inducing the expression patterns of Px genes to convert from D to R was " recessive epistasis" ; (4) the change of combination states of two-linked genes during the expression processes may induce the changes in genetic distance between them in consequence.

本试验结果表明,在玉米萌发胚中,(1)Px基因的表达过程与玉米种胚萌发过程是一致的,其表达顺序和表达频率均受控于遗传调节;(2)Px基因有稳定表达型和可变表达型之分,其中可变表达型Px基因的表达模式能够在一定的遗传背景中以特定的“单基因转换体”和“双基因转换体”的形式发生特定方向的遗传转换;(3)Px基因表达模式发生D→R转换的原因之一是“隐性上位”;(4)两对连锁基因表达过程中结合状态的改变可以引起它们之间的遗传距离随之改变。

正常成人外周血细胞经二步法培养,可得到90%以上纯度的红系细胞。并且红系细胞随培养时间延长,不断向成熟分化,红系细胞数目增多。以逆转录多聚酶链反应法检测该培养体系内珠蛋白链mRNA相对含量。结果表明:随着红系细胞成熟分化,细胞内β-mRNA表达升高,而γ-mRNA表达下降。表明二步法建立的红系细胞液体培养较好地模拟了红系细胞发育的生理状态,为体外研究珠蛋白基因转换提供了良好模型。
